Objective: To observe the effects of Transient receptor potential(TRP)channels in stretch-induced injury of Human brain microvascularendothelial cells(HBMEC) and to explore its mechanism of signalingpathway.Methods: After mechanical stretch by Mini-type multi-functionalbio-impact machine, cells were divided into control group, stretch group,stretch and SKF-96365group, stretch and low molecular weightheparin(LMWH) group and stretch and N-nitro-l-arginine methylester(L-NAME) group. The concentration of free calcium ion in cellsbefore and after mechanical stretch were observed by confocal laserscanning microscopy and determined by flow cytometry. The expression ofeNOS protein was detected by Western blot.Results: The concentration of free calcium ion increased in the role ofmechanical stretch(P<0.05). The expression of eNOS protein increased inthe role of mechanical stretch(P<0.05). Conclusion: Mechanical stretch could increase the concentration offree calcium in HBMEC and the expression of eNOS protein. Themechanism maybe concerned with the activation of Transient ReceptorPotential channels and the influx of calcium.
