OBJECTIVE:To discuss the effect of GubenFangxiao Decoction (GBFXD)on signal transduction and transcriptional activation factor1(STAT1) and interleukin5(IL-5)in a murine model of asthma remission.METHODS:The asthma model was reproduced in the Balb/C female mice challenged by repeated inhalation of ovalbumin(OVA) and repeated intranasal instillation of RS V,montelukast acetate as controlled drugs.Randomly divided into normal group, model group, prevention group,GBFXD group, montelukast acetate group (each group of15). Samples for inspection at the end of the dosing after24hours. Observe the general activities of each group, observe the lung tissue pathological slices by ematoxylin and eosin (HE) staining, Real-time quantitative polymerase chain reaction(RT-PCR) and Western blot assay were employed to confirm the expression of STAT1and IL-5in the lungs of murine model at mRNA and protein levels, respectively. Finally analysis and processing by using statistical methods.RESULTS:1. In normal group, lung tissue of mice structure is clear,no inflammatory exudates appeared in alveolar cavities and the surrounding, the control group, Gas pipe wall and alveolar wall apperred edema and thickening, inflammatory cells infiltration was around, especially the eosinophils. In the prevention,GBFXD and montelukast acetate group, congestive edema were relieved in bronchial wall and alveolar walls, tube wall thickness was reduced, and inflammatory cells infiltration was also significantly reduced, especially the eosinophils.2. Compared with the control group, the expression of STAT1, IL-5were significantly higher in the model group (P<0.01). Compared with the model group, the prevention group,GBFXD group, and montelukast acetate group significantly reduced the expression of STAT1, IL-5, especially in montelukast acetate group,they had differences in statistics.CONCLUSION:1.Excessive expression of STAT1happened in the genesis and development of the asthma.2.To some extent,GBFXD can downregulate the expression of STAT1、IL-5in allergic asthmatic mice, reduce airway inflammatory cells infiltration and prevent the occurrence of asthma.
