| Objective:To investigate the effect of Rhodiola on severe acute pancreatitis associated renal injury and explore the possible mechanisms.Methods:A total of90rats were randomly divided into the control group (S group,n=18), severe acute panceratitis with renal injury group(M group,n=18), low dose group(3.0g/kg)(RHO1group, n=18),middle dose group(6.0g/kg)(RHO2group, n=18),high dose group(9.0g/kg)(RHO3group, n=18). The C and M groups were given6.0g/kg saline through intraperitoneal injection before operation while the RHO1group,RHO2group,RHO3group were given with3.0g/kg,6.0g/kg,9.0g/kg dose of Rhodiola injecta through intraperitoneal injection.The pancreas and pancreatic tail-segment was dissociated and the head of pancreas was occlused in rats to form the model, blood vessel forceps was loosed after three hours. Then take the pancreatic tissue back to home position and close the abdomen to establish the models.All the rats were sacrificed at12h,24h,36h after modeling. The level of ascites,serum amylase, creatinine, blood urea nitrogen, interleukin1β (IL-1β), interleukin6(IL-6)and interleukin10(IL-10) were detected and the pathological chang of pancreas and the left kidney was observed under light microscope,immunohistocheical method was used to detect the expression of HIF-1α protein level in the kidney. The serum levels of IL-1β,IL-6, IL-10was detected by enzyme-linked immunosorbert assay (ELISA).Take the right kidney for Superoxide Dismutase(SOD) determination and the expression of hypoxia inducible factor-1α(HIF-1α) mRNA in the right kidney was detected with real time polymerase chain reaction (RT-PCR). Results:Compared with the sham operated group,the level of serum amylase, creatinine, blood urea nitrogen and IL-1β,IL-6in M group has a significantly increased, but the activity of Superoxide Dismutase (SOD)has a significant decline(P<0.05). RH02group compared with M group,the level of serum amylase, creatinine, blood urea nitrogen and IL-1β,IL-6in RH02group has a significant decline.but the activity of SOD, the express of hypoxia inducible factor-la mRNA and IL-10has a significant increased (P<0.05).RHO1group compared with RH02group,with the dose of Rhodiola increased, the function of the RH02group’s kidney and pancreas has a better performance, the difference has statistical significance (P<0.05).But RH03group compared with RH02group,with the dose of Rhodiola further inceased,the function of rats has not a better improvement, the difference has not statistical significance (P>0.05). Conclusion:Middle dose of Rhodiola(6g/kg) has a good protective effect on severe acute pancreatitis associated renal injury.It may be through inhibited the expression of IL-1β,IL-6, up-regulation expression of IL-10,HIF-la mRNA,and increased the activity of superoxidase dismutase.So it can inhibit the expression of TGF-β and keep the cascade signal pathway of caspases-3off acting,then reduce the cell apoptosis and necrosis of the kidney, improve the ability of the renal cells to tolerate hypoxia. |
