Objective:To study etomidate for rats by Morris water maze spatial learning and memory abilities and using whole-cell patch-clamp combined with pharmacological approaches to long-term depression (Long-term depression, LTD) as an indicator to study etomidate ester on spatial learning and memory ability and hippocampal CA1area suppression effects.Methods:Rats were given etomidate (5mg/kg) by intraperitoneal injection as the experimental group received intraperitoneal injection of fat emulsion used as a solvent control group received saline intraperitoneally as a control group, and then apply the Morris water maze evaluation of etomidate on large spatial learning and memory in rats. After the application of whole-cell voltage-clamp technique was30μl etomidate (equivalent5μmol/L) fat emulsion on rat hippocampal CA1neurons LTD impact and possible mechanisms of etomidate relying impact of LTD.Results:The rats were given intraperitoneal injection of etomidate can affect spatial learning and memory in rats. Etomidate to induce LTD success and LTD increased expression; facilitatory effect of LTD could be competitive GABAA receptor antagonist bicuculline blocked.Conclusion:This study showed that repeated intraperitoneal injection to rats etomidate repeated anesthesia or sedation may affect spatial learning and memory, and the anesthetic etomidate can enhance the expression of LTD, and its mechanism may affect the ability of spatial learning and memory in rats.
