Objective:Laryngeal cancer is a common head and neck malignant tumor,nearly40years, the incidence of laryngeal cancer has a tendency to increase gradually. Laryngeal cancer symptoms mainly for hoarseness, difficulty breathing, repeatedly cough, difficulty swallowing, discomfort, etc., of which were common, and more for starting symptoms. Laryngeal cancer occurs there is currently no known etiology, exactly may is the combined action of many factors lead to disease.A disintegrin and metalloproteinases (ADAMs) is a newly discovered I transmembrane protein family in recent years,because it contains two main structure of the matrix metalloproteinases and disintegrin area.In recent years,members of the ADAM family of proteins,an MMP-related metalloproteinase family,have attracted attention,and functional analyses of ADAMs are ongoing.This article is to study the expression of a disintegrin and metalloprotease10(ADAM10) in laryngeal cancer and its clinical significance.Methods:1.Collect the specimens of total laryngectomy or partial laryngectomy in Department of Otorhinolaryngology, Eye Ear Nose&Throat Hospital of Fudan University between January2011and April2011,a total of21cases.Each specimen in tumor tissue and tumor around5cm above the normal laryngeal mucosa tissue,all specimens in vitro after within10minutes built-in liquid nitrogen preservation to extract RNA.Tissue chip used specimens collected the total laryngectomy or partial laryngectomy of62cases in Department of Otorhinolaryngology, Eye Ear Nose&Throat Hospital of Fudan University in February2010to April2011. Each specimen in tumor tissue and tumor around5cm above the normal laryngeal mucosa tissue,specimens with10%formalin fixed, paraffin embedding to tissue microarray.2.The expressions of ADAM10in the tissue of laryngeal cancer and laryngeal normal mucosa were detected with RT-PCR and tissue chip. The differences of ADAM10expressions between them were analyzed. Results:real-time RT-PCR and tissue chip results showed that the expression of ADAM10in laryngeal cancer was significantly higher than that in laryngeal normal mucosa(P<0.001).The expression of ADAM10in pathological grading group I were significantly different from those in group Ⅱ (P<0.01). However, from the clinical data we found no correlation with staging, typing and metastasis of lymph nodes.Conclusion:The overexpression of ADAM10in laryngeal cancer suggests that ADAM10correlates with the progress and prognosis of laryngeal cancer.
