Relationship Between HO-1and Kidney Injury On Renal In Brain Death Rats

Objective:In the study we establish the brain death model in rats by increasing intracranial pressure (ICP), investigate the correlation between the variation in content of HO-1and the degree of renal damage under state of brain death, probe into the theoretical basis of improving and maintaining the quality of brain dead donor organs.Methods:Total of50adult male rats (weight:350-400g) were divided into5groups randomly. Group A(control group) only give craniotomy,while placing intracranial fogarty3F balloon catheter,without setting up the model of brain death.Group B(0h after brain dead) take specimen immediately after brain dead.Group C、D、E(2h、4h、6h after brain dead) individual take specimen after brain dead2h、4h、6h which was supporting by respirator and circulation.the levels of BUN and Scr in serum were determined by automatic biochemical analyzer,the change of HO-1in serum among the groups was checked with ELISA,Each left kidney samples of rats were harvested for histopathology, PCR to detect the expression level of HO-1mRNA in kidney,Western Blot to detect the expression level of HO-1in kidney.Results:In brain death group, the level of BUN and Scr in serum were actually increases than the levels in control group. Compare with the same time of control group, the diversity had significant difference (P<0.05).Renal histopathological show that renal cell degeneration, mostly in proximal convoluted tubule, and as the extension of induction time the change were more obviously.The level of HO-1in serum and nephridial tissue were higher than the levels in control group, the diversity had significant difference (P<0.05). As the brain dead time extended,The level of HO-1in serum and nephridial tissue was increased firstly and then decreased,But it was still higher than the control group.Conclusion:The content of HO-1in serum and the expression level of HO-1in renal tissue was increased firstly and then decreased,which concluded that the brain dead renal function decreased and traumatic change in the kidney tissue is closely related to the reduced expression of HO-1.
