Objective:The aim of this study was to investigate the effect of hyaluronic acid (HA) on the expression of osteopontin (OPN) mRNA and CD44mRNA of chondrocytes in the patients with osteoarthritis (OA) of the knee.Methods:The study was performed on specimens of cartilage obtained from osteoarthritic patients who underwent knee arthoplasty or arthroscopic surgery. Then all the cartilage samples were cultured and purified to aquire chondrocytes in vitro, and cells are taken into three groups:(1) the control group,(2) treated with100mg/ml hyaluronic acid (HA) for48h,(3) treated with200mg/ml hyaluronidase (HYL) for48h, then the real-time quantitative polymerase chain reaction (Q-PCR) assays designed and validated with SYBR Green dyes for simultaneous quantification of the expression of the CD44mRNA and OPN mRNA. All results statistics were analysed by SPSS17.0.Results:1. There is a statistically significant difference of OPN mRNA expression between each two groups.1) Compared to controls, HA (100ug/ml) upregulated OPN mRNA expression in OA chondrocytes;2) Compared to controls, HYL (200ug/ml) reduced OPN mRNA expression in OA chondrocytes;3) Compared to HYL (200ug/ml) group, the expression of OPN mRNA in HA (100ug/ml) group is higher. 2. There is a statistically significant difference of CD44mRNA expression between each two groups.1) Compared to controls, HA (100ug/ml) reduced CD44mRNA expression in OA chondrocytes;2) Compared to controls, HYL (200ug/ml) reduced CD44mRNA expression in OA chondrocytes;3) Compared to HYL (200ug/ml) group, the expression of OPN mRNA in HA (100ug/ml) group is lower.Conclusion:1HA can upregulate OPN mRNA expression in OA chondrocytes;2The effects of HA on CD44mRNA expression in OA chondrocytes might be associated with the molecule weight of HA.
