Objective: To explore the biological characteristics of bone marrowderived neural stem cells. Characteristics and differences of NSCs derivedfrom different tissues were compared to provide a reliable basis for celltransplantation in clinical diseases.Methods: Bone marrow derived neural stem cells’ characteristics wereexplored by GFP-Nestin mice,including its nestin expression and neuraldifferentiation ability. Immunofluorescence detected the expression ofCXCR4and CD133. Mobilizion of neural stem cells by G-CSF wasobserved. Immunofluorescence was used to show expression of CXCR4and CD133of bone marrow derived neural stem cells. Then, brainderived neural stem cells from fetal mice aged from12ds to15ds wascultured and bone marrow fibroblast cell lines(C3H10)was induced.Differences of neural differentiation in bone marrow fibroblasts derivedNSCs、bone marrow mononuclear derived NSCs and brain derived NSCs were identified by immunocytochemistry. RT-PCR were performed todetect the expressions of mRNA including stem cell-related genesNotch1、Nanog、Emx2and cycle-related genes CyclinD1、CDK4、CyclinB1、CDC2.Results: Bone marrow mononuclear cells from the mice model containa group of cells which have the characteristic of neural stem cells、Nestinexpression ability and the potential of neural differentiation. The number ofbone marrow derived neural stem cells was significantly increased (P <0.05)under the mobilization of G-CSF.Different sources of stem cells can beinduced to forming neurospheres, expressing Nestin of NSCs. All kinds ofNSCs had|-Ⅲ-tubulin、GFAP positive cells after differentiation and therewere differences in proportion. The results of mRNA detection showed thatdifferent sources of stem cells expression stem cell-related genes Notch1、Nanog and cycle-related genes CyclinD1、 CDK4、CyclinB1、 CDC2afterinduction. However there are some differences among the groups. Fetalbrain and bone marrow fibroblasts derived NSCs had almost no expressionof Emx2.However, there was a high expression in mononuclear cells drivedNSCs.Conclusion: Different sources of adult stem cells have characteristicsof neural stem cells after induction.However, there are differences in theproliferation and differentiation and even the gene expression in differentsources of NSCs. Bone marrow mononuclear cells is a good source of NSCs.But the low proliferative rate is a big limitations and the cultureconditions may need to be further improved. Bone marrow fibroblastsderived NSCs had the characteristics of rapid proliferation, good spheresforming ability, higher rate of differentiation and more stem cellcharacteristics after induction.It may be one of the most advantage adultstem cells derived NSCs used to replace brain-derived NSCs applicating inclinical treatment.
