[Objectiv] Make research on the quality standards of Tibetan medicine commonlyused "Dida"(Zang Yin Cheng), establish quality control and evaluation methods ofSwertia and Indian Swertia herbs, and to develop quality standards of of Swertia andIndian Swertia herbs, so as to provide reference and basis for quality control andresearch of Swertia and Indian Swertia herbs.[Methods](1) In the "source", according to the study results such as textualresearch, investigation of resources and the status of using, component analysis andpharmacodynamics, we set the original plant collected in each former standard as thesource of the same medicinal variety;(2) Pharmacognosy research of Swertia and Indian Swertia herbs from the aspectsof plant morphological characteristics, medicinal properties, microstructure and so on;(3) Through systematic chemical components separation, we made systematicstudy on the chemical compositions of swertia herbs and synchronously made analysison the chemical compositions of Indian Swertia;(4) Moisture content and ash content research of Swertia and Indian Swertia herbsbased on the2010edition of "Chinese Pharmacopoeia"(5) Qualitative identification of Swertia and Indian Swertia by TLC(6) Content determination of Mangiferin, sweroside or swertiamarin glycoside inSwertia and Indian Swertia by HPLC.(7) Formulate a draft medicinal quality standards of Swertia and Indian Swertiareferring to the2010edition of "Chinese Pharmacopoeia" and make comparison withthe original ministerial standards.[Results] In order to improve and enhance the quality standards of Swertia and Indian Swertia,this study has completed the following tasks:(1) Identification of PharmacognosyAccording to examination of tissue sections(2) The results of the chemical compositions study showed that both Swertia andIndian Swertia mainly contain chemical compositions such as iridoid, triterpenoidsaponins, San ketones and polysaccharide. By separating the parts of chloroform andn-butanol, we got16compounds from Swertia altogether(3) Considering the influence of various factors, we recommend that the moisturecontent of Swertia is not more than13.0percent, and ash content is not more than5.5percent, while the moisture content of Indian Swertia is not more than14.0percent, andash content is not more than4.0percent.(4) TLC method showed good separation, clear spots, good durability, and goodreproducibility.(5) Method for the determination showed high separation, good durability, goodaccuracy, and also good reproducibility[Conclusions] This study improved the standards of Swertia and Indian Swertia asfollows:(1) Increasd the "stem cross-section" in Identification;“TCL” controlled byoleanolic acid;(2) Accordance with the first method of moisture content determinationin the "Pharmacopoeia" Appendix IX H, and the method of ash content determination inthe Appendix IX K, we increased limited regulations of moisture content and total ashcontent in the project of Check;(3) Increased the content limits formulation in theproject of Determination of content by HPLC, Swertia uses swertiamarin glycosides andMangiferin as indicators,Indian Swertia uses Sweroside and Mangiferin as indicators.And formulated the draft quality standards of Swertia and Indian Swertia by referenceto the2010edition of "Chinese Pharmacopoeia", this standard has scientific rationality,high accuracy and practical, and provides a reference for the quality control of Swertiaand Indian Swertia.
