Objective: To study the effect of the TW in the osteogenicdifferentiation of rabbit bone marrow mesenchymal stem cells by theRT-PCR and SEM. The experiment is desinged to provide a good scaffoldmaterial for bone defect repair restructuring.Methods: The rabbit bone marrow mesenchymal stem cells wereisolated and cultured with whole bone marrow adherent culture method byusing Percoll Pharmacia. The3thgenerations were chosen to be inducedinto osteoblasts and lipoblasts to prove the ability of multi-directionaldifferentiation of BMSCs. The3thgenerations were obtained and culturedin the Six orifice plate. The TW and ordinary culture medium were used inthe experiment group; the osteogenic differentiation inducer with ordinaryculture medium was used in the control group; only ordinary culturemedium was in the blank group. After14d following culture, RT-PCR wasutilized to determine expression of ALP、OPN、OCN. The scanningelectron microscope was used to observe the rabbit bone marrow mesenchymal stem cells adhering and growing into the TW and the cellularmorphology.Result: RT-PCR results showed that the expression of ALP、OPN、OCN was higher than the control group and blank group in the experimentgroup. The scanning electron microscope results showed that the rabbitbone marrow mesenchymal stem cells can adhere to the TW, grow into theTW. The cellular morphology was polygon or spindle, keep themorphological specificity of stem cells.Conclusion: TW can Successfully and significantly induce theosteogenic differentiation of rabbit bone marrow mesenchymal stem cells,so it can be the ideal support material in the bone defect repair.
