| Objective:To investigate the relationship between EZH2and age-relatedislet cell function.Methods: Healthy SD rats were divided into4groups by age:1,6,12,24months of age group respectively,6rats in each group,Usingimmunohistochemistry methods, expression level and location of EZH2were detected originally,simultaneous detection of islet cellproliferation,apoptosis;Then western blot was performed to determineaccurate expression level of EZH2、P16and insulin. At last, we analyzedEZH2and age-related islet cell function relationship.Results:With age increasing,the expression of insulin in the isletdecreased,the average gary level of islets respectively:1months of agegroup206.5±27.8；6months of age group267.4±25.3；12months of agegroup376.2±31.9；24months of age group187.8±25.1（P<0．05）.theexpression of EZH2in islet was gradually decreased,the number of EZH2positive cells-to-islet total nucleated cells ration:1months of age group0.22516±0.03091；6months of age group0.18427±0.02315；12months of age group0.03165±0.01675；24months of age group0（P<0．05）.proliferation of islet cells were also gradually decreased,thepercentage of positive cells proliferation:1months of age group13.43±2.02%；6months of age group10.78±1.58%；12months of agegroup6.23±0.78%；24months of age group2.10±0.53%（P<0．05).apoptosisof islet cells were gradually increased,the percentage of apoptosis positivecells:1months of age group2.53±1.56%；6months of age group12.34±2.01%；12months of age group20.06±2.34%；24months of agegroup27.72±2.84%（P<0．05).Conclusions:With increasing age, the islet cells were gradually aging,theexpression of EZH2was gradually reduced in aging islet cells,accompanied by decreased islet cell proliferation, apoptosis, and insulinlevels gradually reduced. |
