Objective:Recurrent spontaneous abortion (RSA) is the occurrence of three or more early abortion. The pathogen of RSA is not fully understood. The purpose of this study is to investigate the expression and correlation of Survivin, Bcl-2and Bax in chorionic villi of recurrent spontaneous abortion patients. Methods:human chorionic villi of RSA patients and artificial abortion patients were collected. The expression of Survivin, Bcl-2, and Bax in chorionic tissues was detected by immunohistochemistry staining. The positive incidences were calculated by cell counting. Apoptosis of chorionic tissues were detected by Terminal-deoxynucleoitidyl Transferase Mediated Nick End Labeling (TUNEL). The protein and mRNA expressions of Survivin, Bcl-2, and Bax in chorionic villi were detected by western blotting and real time PCR respectively. The correlation between Survivin, Bcl-2and Bax were analyzed using Cox regression. Results: Immunohistochemistry staining showed that the positive of Survivin, Bcl-2, and Bax in RSA group were44.59±7.37%,51.46±6.33%and86.73±10.92%respectively. The positive of Survivin, Bcl-2, and Bax in normal group were85.36±8.64%、78.32±7.51%and20.05±4.65%respectively. The apoptotic index (AI) of RSA and normal group were49.57±6.81%'8.26±2.13%respectively. Both western blotting and real time PCR data showed that the expression of Bcl-2and Survivin is downregulated in RSA, while the expression of Bax is up-regulated. There was a positive correlation between the expression of Survivin and Bcl-2(r=0.40, p<0.05), whereas a negative correlation between the expression of Survivin and Bax (r=-0.56, p<0.05). Conclusion:Survivin, Bcl-2and Bax genes may play an important role in the pathogenesis and development of RSA. Survivin and Bcl-2, bax have a synergic or antagonistic role respectively during the RSA.
