| Objective:To investigate the expressions of Shh,Ptchl,Smo and Glil in the pancreatic cancer and the pancreatic cancer adjacent tissue, We would expect to explore the molecular mechanism of pancreatic cancer. Methods:RT-PCR and Western blot were used to determine the mRNA and protein expression of Shh,Ptchl,Smo and Glil in50cases of pancreatic cancer and pancreatic cancer adjacent tissue.The relationship between the expression of Shh,Ptchl,Smo and Glil and the clinicopathological features of pancreatic cancer was analyzed.Results:The expression of Shh,Ptchl,Smo and Glil mRNA in pancreatic cancer was0.309±0.162,0.413±0.115,0.327±0.264and0.341±0.132, and in pancreatic cancer adjacent tissue was0.184±0.227,0.203±0.143,0.148±0.276and0.105±0.351by RT-PCR detected(P<0.05).The expression of Shh,Ptchl,Smo and Glil protein in pancreatic cancer was0.624±0.139,0.724±0.201,0.547±0.418and0.563±0.271,and in pancreatic cancer adjacent tissue was0.388±0.294,0.516±0.198,0.294±0.152and0.328±0.129by Western blot detected(P<0.05).The expression of Shh,Ptchl,Smo and Glil mRNA and protein in pancreatic cancer tissue were both higher than pancreatic cancer adjacent tissue.Overexpression of them were concerned with differentiation of pancreatic cancer(P<0.05),not age, tumorous size,TNM grade,invasion to nerves and vascellum,lymph node metastasis,distant metastasis(P>0.05). Conclusion: The expression level of Shh,Ptchl,Smo and Glilsignal molecule is increased in human pancreatic cancer(hPC).The overexpression of hedgehog signaling pathway might play an important role in carcinogenesis and development of pancreatic cancer. |
