With the aging of the population, the incidence of perimenopause syndrome increases as well. The dysfunction of neuroendocrine immune function is seen as the basic of perimenopause syndrome. At the present the main therapeutic method in domestic and overseas is hormone replacement therapy, but the side effect and contraindication limit its clinical application. Traditional Chinese medicine therapy has been the main therapeutic method in curing perimenopause syndrome because of the small side effect and remarkable effects. GNC which is researched according to Chinese medicine theory and the clinical experience of many years has acquired favorable curative effect in improving perimenopause syndrome. So it is essential to study the treat mechanism of GNC for treating perimenopause syndrome. This article is to observe the effect of GNC on neuroendocrine immune functions in ovariectomized rats, to explore the mechanism of GNC against perimenopause syndrome, to provide the scientific basis for clinic application.Part Ⅰ:Effect of Herbal Formula GengNianChun on Learning and Memory Ability in Ovariectomized RatsObject:Observe the effect of GNC on learning and memory ability in ovariectomized rats, to explore the mechanism of GNC in nervous system.Methods:There were7groups in this experiment. Three months old SD rats were randomized into GNC group (OVX+GNC group), estradiol valerate group (OVX+E2group) and normal saline group (OVX+NS group) after ovariectomy. The rats were randomized into GNC group (Sham+GNC group) and normal saline group (Sham+NS group) after excising adipose tissue surrounding double ovaries. The rats without any operation were randomized into normal saline group (NC+NS group) and normal control group (NC group). This experiment is to test the learning and memory of ovariectomized rats after3-month gavage by water maze experiment.Results:(1) In the place navigation performance of Morris water maze, the escape latent periods in OVX+NS group were longer compared with that of NC+NS group, NC group (P<0.05); The escape latent periods in OVX+GNC group, OVX+E2group were shorter compared with that of OVX+NS group (P<0.05); The escape latent periods in Sham+GNC group were longer compared with that of Sham+NS group%NC group (P<0.05). There was no obvious difference between OVX+E2group and OVX+GNC group in the escape latent periods.(2) In the spatial probe performance of Morris water maze, the times across platform in OVX+NS group were decreased compared with NC+NS group and NC group (P<0.05); the times across platform in OVX+GNC group, OVX+E2group were raised compared with OVX+NS group (P<0.05); the times across platform in Sham+GNC group, Sham+NS group were decreased compared with NC group (P<0.05); There was no obvious difference between OVX+E2group and OVX+GNC group in the the times across platform. In the spatial probe performance of Morris water maze, the trail in OVX+NS group distributed randomly compared with NC+NS group and NC group, the trail in OVX+GNC group、OVX+E2group concentrated in platform compared with OVX+NS group. The trail in Sham+GNC group与Sham+NS group distributed randomly compared with NC group.Conclusion:Castration will decline the learning and memory ability; GNC can improve the learning and memory ability in ovariectomized rats, and the effect is similar to estrogen; uncastrated rats with GNC will decline the learning and memory ability.Part Ⅱ:Effect of Herbal Formula GengNianChun on Neurotransmitters, Cytokines, Leptin and Endometrial Thickness in Ovariectomized RatsObject:Observe the effect of herbal formula GengNianChun on neuroendocrine immune functions in ovariectomized rats, to explore the mechanism of GNC against perimenopause syndrome. Methods:There were8groups in this experiment. Three months old SD rats were randomized into high-dose GNC group (OVX+GNCH group), middle-dose GNC group (OVX+GNCM group), low-dose GNC group (OVX+GNCL group), high-dose estradiol valerate group (OVX+E group), low-dose estradiol valerate group (OVX+E group), double distilled water group (OVX+D group) after ovariectomy. The rats were randomized into middle-dose GNC group (Sham+GNCM group) and double distilled water group (Sham+D group). These rats were fed for30days respectively. The weight; the content of hypothalamic neurotransmitters such as norepinephrine, dopamine,5-hydroxy tryptamine,5-hydroxyindoleacetic acid; the content of estrogen; the content of IL-1β、IL-6、TNF-α、IL-2; the content of leptin in rat serum, and the endometrial thickness were measured.Results:(1) The level of NE, DA in the hypothalamus of rats in OVX+D group was markedly decreased and the level of5-HT,5-HIAA,5-HT/NE,5-HT/DA was markedly raised compared with the rats in each group of Sham groups (P<0.05). The level of DA in the hypothalamus of rats in each group of OVX+GNC groups and each group of OVX+E groups was markedly raised and the level of5-HT,5-HIAA,5-HT/NE,5-HT/DA was markedly decreased compared with the rats in OVX+D group (P<0.05). There was no obvious difference among each group of OVX+GNC groups and OVX+E group in the level of hypothalamic neurotransmitters. The level of5-HT,5-HT/NE,5-HT/DA in the hypothalamus of rats in OVX+GNCH group、OVX+EH group was markedly decreased compared with OVX+GNCM group、OVX+GNCL group(P<0.05).(2) The level of serum IL-1β、IL-6、TNF-a in OVX+D group was markedly raised compared with the rats in each group of Sham groups (P<0.05); The level of serum IL-2、 IFN-y in OVX+D group was markedly decreased compared with the rats in each group of Sham groups (P<0.05); The level of serum IL-1β、IL-6、TNF-a in Each group of OVX+GNC groups, each group of OVX+E groups was markedly decreased compared with the rats in OVX+D group(P<0.05); The level of serum IL-2、IFN-γ in each group of OVX+GNC groups, each group of OVX+E groups was markedly raised compared with the rats in OVX+D group (P<0.05). There was no obvious difference among each group of OVX+GNC groups and OVX+E group in the level of serum cytokines. The level of serum IFN-γ in OVX+GNCH group was markedly raised compared with the rats in OVX+GNCL group (P<0.05). The level of serum IFN-γ in OVX+EH group was markedly raised compared with the rats in OVX+EL group (P<0.05).(3) There was no obvious difference among each group before gavage. After30-days gavage the weight and the level of leptin in OVX+D group was marked raised compared with rats in each group of Sham groups (P<0.05); the weight and the level of leptin in each group of OVX+GNC groups and each group of OVX+E group was marked decreased compared with rats in OVX+D group (P<0.05). There was no obvious difference among each group of OVX+GNC groups and OVX+E group in the weight and the level of leptin.(4) The endometrial thickness in OVX+D group was more thinning compared with that in Sham+GNCM group, Sham+D group (P<0.01); the level of estrogen in serum in in OVX+D group was decreased compared with that in Sham+GNCM group, Sham+D group (P<0.01); There was no obvious difference among each group of OVX+GNC groups and OVX+D group in the endometrial thickness and the level of estrogen; the endometrial thickness and the level of estrogen in each group of OVX+E groups were raised compared with OVX+D group and each group of OVX+GNC group (P<0.01).Conclusion:GNC can regulate the secretion of hypothalamic neurotransmitters, improve the immune function of ovariectomized rats, regulate the lipid metabolism, but not improve the endometrial thickness.
