Injection stauntoniae which is one of chinese traditional medicines can be used torelieve pain.The purpose of our present work is to study the effects of injectionstauntoniae which is consisted of effective components from stauntoniae on transientpotential vanilloid receptor1(TRPV1)currents in dorsal root ganglion neurons andanalyze its pharmacological mechanism of blocking the nerve conduction andanalgetic action. Whole-cell patch-clamp recordings were performed in acute-isolatedrat dorsal root ganglion neurons and the effects of1%,5%,10%,15%,25%,50%,75%and80%injection stauntoniae on TRPV1currents were observed. In the absenceand presence of the IS, the standardized capsaicin-induced TRPV1currents were usedas a function of the capsaicin concentration to draw the dose-response curve ofcapsaicin.And we study the injection stauntoniae effects in capsaicin model of pain inmice. Injection stauntoniae inhibited the peak TRPV1currents in dorsal root ganglionneurons in a dose-dependent way.Just increasing the capsaicin concentration couldoffset the inhibition produced by the injection stauntoniae on capsaicin-inducedTRPV1currents, which indicated that the injection stauntoniae may antagonize TRPV1in the competitive antagonistic model. Pain caused by capsaicin can be reduced indose-dependent way in vivo animal experiments. The injection stauntoniae couldmarkedly inhibit capsaicin-evoked currents and block the transmission of painsensation,which may contribute to one of analgesic mechanisms of stauntoniae.
