BAFF is mainly expressed both on neutrophilic granulocyte and macrophage. There was a little expression could be seen on the actived T lymphocyte cell and dendritic cell. B-lymphocyte activating factor (BAFF) is a key survival factor for B-cells and ensures their existence through reducing apoptotic clearance.An elevated serum level of TNFa is associated with the pathophysiology of rheumatoid arthritis, systemic lupus erythematosus ankylosing spondylitis, etc, and neutralization of BAFF has become a new strategy in the therapy of these diseases. So our research have focused on developing BAFF antagonistic peptides that could be prepared synthetically and administered at high concentrations. Peptides sequences are designed based on the crystal structure of the complex of soluble human BAFF receptor with human BAFF, then construction expression and activity analysis of TA/TC-Fc proteins. These peptibodies had highly affinity with BAFF and inhibited the B cell proliferation and which could alleviate the symptoms of rheumatoid joint model in mice, could be a potential biological agents for the treatment of autoimmune diseases. |
