Objectives The effects of astragalus polysaccharide will be explored on immunefunction of organism in vivo and in vitro experiments, which provide experimentalevidence for clinical application and related product development.Methods This study is divided into two parts, in vivo and in vitro experiments. In vivoexperiment, fourty SPF Kunming mice(half male and female,4~6weeks, weight20±2g)were randomly divided into four groups, APSⅠgroup (low dose group of astragaluspolysaccharides), APS Ⅱ group (middle dose group of astragalus polysaccharides), APSⅢ group (high dose group of astragalus polysaccharides) and the NC group (normalcontrol group), and10mice in each group. The APSⅠ, Ⅱ, Ⅲ groups were fed with0.2ml astragalus polysaccharides solution of concentration of5mg/ml,10mg/ml,20mg/ml respectively every day, NC group with0.2ml normal saline every day. After10days feeding, the mice were killed without pain and the samples were collected from them.The immune function of the mice were examined as follows.(1) The innate immune: themacrophage function was detected;(2) The cellular immune: weight difference of left andright postpedes were checked;(3) The humoral immune: antibody aggregate was studied;(4)The immune system: spleen and thymus indexes were researched. Experiment in vitro:take spleen cells from four mice, and place them in24wells of the cell culture plate, set theAPS group (astragalus polysaccharides group) and the BC group (blank control group).Use WST-1method to detect cell proliferation activity, ELISA method for determinationof cytokines IL-2, IFN-γ and flow cytometry technology to detect cell phenotype.Results Experiment in vivo: Phagocyte rate and phagocyte index of APSⅠ, Ⅱ, Ⅲgroup were significantly higher than that of NC group (P<0.001); Weight difference ofleft and right postpedes of APSⅠ, Ⅱ, Ⅲ group were significantly higher than that of NCgroup (P<0.001); Antibody aggregate of APSⅠ, Ⅱ, Ⅲ group were significantly higherthan that of NC group (P<0.001); Spleen and thymus index of APSⅠ, Ⅱ, Ⅲ groupwere significantly higher than that of NC group (P<0.001). Experiment in vitro: APSgroup had a significantly higher cell proliferation activity than that of BC group (P<0.001); The concentration of IL-2, IFN-γ of APS group, was obviously higher than thatof NC group (P<0.001), and increased with the increase of the concentration ofastragalus polysaccharides, with a dose-response relationship within a certain concentration range;The expression of CD3, CD19cell of APS group were higher than thatof BC group (P<0.001).Conclusions (1) Astragalus polysaccharide can significantly improve the immunefunction of organism, including natural immune function, cellular immune function,humoral immune function.(2)Astragalus polysaccharide can not only enhancethe humoral immune function, but also enhance the cellular immune function of micebody, changed people usually think that astragalus polysaccharide can enhance only thehumoral immunity function.
