Objectives This experiment by using immunohistochemical and real-time PCR methodto detect breast infiltrating ductal carcinoma and mammary gland hyperplastic lesions ofNF-kB, VEGF-C and the expression of VEGFR-3levels, provide relevant molecularpathology for the prognosis of breast cancer detection.Methods78cases of breast invasive ductal carcinoma,20cases of mammaryhyperplasia were collected from the file samples of the affiliated hospital of Hebei UnionUniversity during Jan.2009to May.2013.Using immunohistochemical SP method ofdetection of these organizations NF-kB, the expression of VEGF-C and VEGFR-3protein; Real-time PCR method was employed to detect the mRNA expression of NF-kB、VEGF-C and VEGFR-3in20cases of breast invasive ductal carcinoma,15cases ofmammary hyperplasia.Results1)immunohistochemical detection results:(1)The NF-kB, VEGF-C andVEGFR-3in breast invasive ductal carcinoma group, the positive expression rate ofgroup were significantly higher than that of hyperplasia of mammary glands, statisticallysignificant difference (P<0.05);(2)The NF-kB, VEGF-C and VEGFR-3in breastinvasive ductal carcinoma pathological grading Ⅲlevel organization levels significantlybetter than the positive rate of Ⅰa nd Ⅱ, statistically significant difference (P<0.05);(3)The NF-kB, VEGF-C and VEGFR-3protein with lymph node metastasis in breastinvasive ductal carcinoma group the positive rate is higher than without lymph nodemetastasis in breast invasive ductal carcinoma group, comparing the two groups havesignificant difference (P<0.05);(4)The NF-kB, VEGF-C and VEGFR-3protein inbreast invasive ductal carcinoma group, the positive rate was significantly related withTNM staging Ⅲ-Ⅳ significantly better than the positive rate ofⅠ-Ⅱ, differencesstatistically significant (respectively, P<0.05);(5)NF-kB、VEGF-C protein in breastinvasive ductal carcinoma tissue expression of has nothing to do with the tumor size andsicken age (P>0.05);(6)VEGFR-3in the lymph node metastasis group positive LVDmean compared and lymph nodes transfer group, significant difference, statisticallysignificant (P<0.01);(7)VEGFR-3in breast invasive ductal carcinoma group, positivelymphatic vessel density (LVD) mean compared with mammary gland hyperplasia group, significant difference, with statistical significance (P<0.01).2)real-time PCR detection results:NF-kB, VEGF-C and VEGFR-3mRNA in breastinvasive ductal carcinoma of the positive expression rate were significantly higher thanthat of breast benign tumor, statistically significant difference (P<0.05). Three relativeexpression and infiltrating ductal carcinoma of the breast is related with lymph nodemetastasis and TNM stages (P<0.05), the difference is statistically significant. The threerelative expression with the patient age, tumor size and histological classification of thedisease by irrelevant (P>0.05).Conclusion1. The experimental results show that the NF-kB, VEGF-C and VEGFR-3are associated with malignant progress of breast invasive ductal carcinoma.2. NF-kBabnormal activation can increase VEGF-C expression in breast invasive cancer cells,then the VEGF-C in the form of paracrine combined with infiltrating ductal carcinomafrom breast cancer tissue lymphatic endothelial cells of VEGFR-3, thus promotes theformation of tumor lymphatic around and lymphatic metastasis.
