There are various types of neurotransmitters with different functions inanimals.Neurotransmitters such as acetylcholine (ACH) and gamma-aminobutyric acid(GABA) have high biological activities and play an important role in regulatingbiological functions of living organisms. Yet there is a lot of evidence thatabnormities of ACH and GABA will lead to many diseases such as AlzheimerDisease. Conclusion the determination of level of ACH, GABA and theirmetabolites in patients are important indexes in evaluation of disease progress anddrug efficacy. For the time being, it is difficult to detect the concentration change ofACH and GABA using living animals. To solve this problem, we collect dialysatesamples using Stereotactic brain surgery and brain Microdialysis techniqueinstrument and detect the concentration change of ACH and GAMA byultra-performance liquid chromatography chromatography tandem massspectrometry (UPLC-MS/MS). In new drug development, various of biologicalsamples can be used for disease diagnosis and drug efficacy evaluation. Various ofsampling methods have been used in sample collection, Microdialysis technique,which is a newly thriving sampling method, is gaining growing attention from allover the world and will pay a larger role in biopharmaceutical research&development.The UPLC-MS method has been established for determining the content ofACh and GABA. Results: The method is proved to be rapid, simple, accurate andsuitable for the ACh and GABA analysis. Methods: The study uses the differentconcentration on the recovery of the probe using incremental and reduction method.Results: The recovery of microdialysis probe was no significant affect on theconcentration of influence, the probe recovery in vivo has good stability. Conclusion:The fully validated UPLC-MS/MS method has been successfully applied todetermination of Ach and GABA in microdialysates samples from rat’s brain.
