MethodsThe relative expression levels of mi R-30 a in the colorectal cancer cells that have different migration abilities were detected by stem-loop reverse transcription quantitative real time polybrerase chain reaction(stem-loop q RT-PCR).mi R-30 a precursor molecules(pre-mi R-30a) were transfected in SW620 to increase the expression levels of mi R-30 a,then implementing the following function tests. Transwell method to detect cell invasion; wound healing method to detect cell metastasis; Western blotting method to detect the expression levels of astrocyte elevatedgene-1(AEG-1). Results1.The relative expression levels of mi R-30 a in HT-29/HCT-116 and SW620 were significantly lower than that in SW480,especially lowest in SW620.2.Biology software analysis show that AEG-1/MTDH is likely the target of mi R-30 a,compared with the control group,the expression level of AEG-1/MTDH m RNA or protein significantly decreased after overexpression of mi R-30 a.
