The Observation Of The Nestin、Sox2-Positive Cells On Median Eminence In Rats Of Different Ages

Objective:To observe the change of the distribution and the morphology of the Nestin,Sox2-positive cells on median eminence in rats of different ages.Methods:Choosing health male- rats of 24,youth rats 12 and adult rats 12,the rats which were anesthetized were primed with 4% paraformaldehyde(0.1mol/L,PH7.2)through the heart.The youth rats and the adult ones randomly divided into two groups seprately,The nestin group(nyouth=6,nadult=6)The sox2 group(nyouth=6,nadult=6):The Nestin,Sox2 on median eminence of the two groups which bothget dehydration,transparention,embeddin,and sliced were detected by immunohistochemical staining.1 Specimens drawn:The normal rats were anesthetized by Chloral hydrate(0.3ml/100g),4% paraformaldehyde(0.1mol/L,PH7.2)was primed through the ascending aorta,fixed,got the brain,then fixed.Choosing the median eminence of the two groups which both getdehydration,transparen-tion,embedding,used the Paraffin slicing machine to get the coronal slice planewhich the thickness of 5μm.Disposal of the experimental animals with the animal ethics requirements.2 Immunohistochemical staining:The Paraffin sections were divided into two,each part were stained with Nestin and Sox2.Then the sections were mounted with neutral gum,taken the pictures with the Olympus BX-UCB.3 Statistical analysis:Calculated the number of the positive cell under the same square which are in the same multiple photos,in the same depth,and the same length of ependymal layer.And calculated the positive rate.Results:1 The results of Immunohistochemical Nestin observed:Nestin positive cells was significantly expressed in the ME Office.In ME ependymal zone and the internal zone of the youth rats,the distribution of the tape showed strong positive.While in ME ependymal zone and the external zone of the adult rats,the distribution of the tape showed strong positive.The protruding portion packed the line from the dorsal to ventral,toward to the soft membrane of the external zone and the vessel wall.Compared with adult rats(the positive expression rate 57.63±4.90%),the youth rats(the positive expression rate 29.81±4.55%)in the total number of positive cells of Nestin.There is a significant difference between two groups(P<0.01).2 The results of Immunohistochemical Sox2 observed:Sox2 immunoreactive cells in ME ependymal with monolayer neat distribution.In the internal zone of the ME of the youth rats,the distribution of the tape showed strong positive.While in the adult rats, the distribution of the tape showed light positive.The positive cells of Sox2 of the youth rats(the positive expression rate:44.30±3.50%) is more than the adult ones(The positive expression rate:31.40±4.03%).There is a significant difference between two groups(P<0.01).Conclusions:1 Nestin immunoreactive cells are mainlydistribute in the ependymal zone,and in the ME internal zone,its’ projections are long and walk the line from the dorsal to ventral,toward to softmembrane of the external zone and the vessel wall,the positive cells of adult rats is more than the youth ones,the expression of intermediate zone is more stronger than the external zone in the youth rats.2 Sox2 immunoreactive cells are densely distributed in the external of the ME ependymal zone.The positive cells in the ME of the adult rats are sparsely distributed.The positive cells of youth rats is more than the adult ones.
