Purpose: To test invastigate the meaning of Tyrosine kinase 2(DDR2) and vascular endothelial growth factor(VEGF) and the correlationship of the two factors by testing the expressions of of the two factors in of I-V class glioma tissues defined by WHO and normal brain tissue.Method: To collect 85 cases of glioma tissues from the second affiliated hospital of hebei medical university----betwee from May in 2013 to October in 2014,there are14 cases of class I, 27 cases of class II, 25 cases of class III,19 cases of class IV.Cases of specimen were diagnosed by the pathological department of hebei university department according to the WHO diagnostic criteria for brain tumor classification. 15 normal brain tissue specimens were taken from patients with traumatic brain injuries or patients with cerebral hemorrhage who needed Intracranial decompression.All the glioma specimens were diagonosed for the first times after admission and had without any anti-tumor treatment such as radiation and chemical therapy before ntracranial decompression operation. All experimental specimens were cut immediately from pathological tissue,and wered fixed by 10% formalin and were stored by liquid nitrogen at- 80 ℃,All the specimens were divided into five groups: normal brain tissue, grade I glioma group, grade II glioma group, grade III glioma group, grade IV glioma group.Use immunohistochemical method to detect VEGF and DDR2 expression in the pathological tissue section of the five groups, use statistical software SPSS13.0 to analyze its relevace, use immunofluorescence to detect the expressions of VEGF and DDR2 and observe the distribution characteristics of the two factors. Use Western Blot method to determine the VEGF and DDR2 protein bands in the specimens of the five groups, and applicate related software to test grey value in the band of the five groups, use SPSS13.0 statistical analysis.Results: Protein expression1 In IHC results showed that the positive rates of VEGF in normal brain tissue, I, II, III, IV grade glioma group expression were 6.67%, 71.43%, 88.89%, 71.43%, 100.00%, and the expression of VEGF increased with the malignant degree of tumor, Spearman rank correlation was concluded and the correlation coefficient r = 0.698, P < 0.01, the expression of VEGF was positively correlated with tumor grade, the higher tumor grade, the higher the positive rate of VEGF expression, as shown in the pictures: Fig1、Fig2、Fig3、Fig4、Fig5,Statistical chart was shown in the Table 1、Table 32 IHC results show: the positiv rates of DDR2 in normal brain tissue, I, II, III, IV grade glioma groups were 33.33%, 78.57%, 88.89%, 78.57%, 100.00%, and the expression of DDR2 increased with the malignant degree of tumor, Spearman rank correlation was concluded and the correlation coefficient r = 0.588, P < 0.01, the expression of DDR2 was positively correlated with tumor grade, the higher tumor grade, the higher the positive rate of DDR2 expression, as were shown in the picture: Fig6、Fig7、Fig8、Fig9、Fig10, see Table 2 and Table 43 Correlation analysis of the expressions of VEGF and DDR2 were conducted. The correlation coefficient r = 0.928, P < 0.01, the expression of VEGF was highly positively correlated with the expression of DDR2, the higher level expression of VEGF, the higher level expession of DDR2, as were shown in the Table 5.4 Using double immunofluorescent staining observation of two kinds of factordistribution of the same pathological tissues. The red fluorescence of VEGF factor, the green fluorescence of DDR2 factor, blue fluorescent nuclear. Visible distribution of two kinds of factors in the same tissues were gathered together. See Fig11, Fig12, Fig13;5 Protein content: use western blot method to determinate the DDR2 and VEGF protein expessions in the specimens of the normal brain tissue, grade I, grade II, III, IVglioma groups, we use the Stripe grey value as standard. Analyse the relative expression of VEGF and DDR2, the higher relative expression of VEGF,the lower relative expression of DDR2. All levels of glioma group had significant differences from that of normal brain tissue in the expressions of the two factors, using the single factor analysis of variance. Results was shown by mean + /- standard deviation, says P < 0.05, As were shown in the Table 6, Fig 14, Fig 15, Fig 16Conclusion:1 DDR2 and VEGF protein contents in glioma tissues were higher than those of nomal tissues,the higher tumor grade, the higher level of DDR2 and VEGF protein contents,and had significant differences between the groups.2 DDR2 and VEGF protein contents in glioma tissues were higher than those of nomal tissues,the higher tumor grade, the higher level of DDR2 and VEGF protein contents,the malignant level can affect the DDR2 protein expression to a certain extent.3 It can Can be seen in the immunofluorescence images that in multiple sets of specimens, the DDR2 protein expression site and VEGF protein expression is coincidence. Combined with statistical analysis, DDR2 and VEGF has certain relevance.4 Through the normal brain tissue and different grade glioma group immunohistochemistry analysis, Spearman rank correlation between P<0.01, VEGF expression were positively correlated with DDR2 expression.
