The Correlation Study Of The Co-expressions Of AR/CD 133 In Glioma Stem Cells And The Analysis Pointing To The Risk Factors In Glioma

Objective To explore the co-expressions of AR/ CD133 in U87 glioma cell line and glioma stem cells,as well as the clinlical significance.To analysis the correlation between the co-expressions of AR/CD133 and the stemness of glioma stem cells.To investigation the risk factors and the community prevention in glioma.Methods(1)Nanosphere suspension culture method combina with neural stem cell medium have been used by us to culture glioma stem cells,which is isolated from U87 glioma cel I line.The expression pattern of AR and CD133 was investigated by Immunofluorescence technique, Real-time PCR and western blot respectively in in U87 glioma cell line and glioma stem cells.(2) The 1:1 matched case-control study method have been used by us.The conditional univariate and multivariate Logistic regressions have been used by us to analyse the risk factors related to cerebral gliomas pathogenesis.Results(1) The relative m RNA level of AR was 1.03±0.11 in U87 glioma cel I line,1.56±0.12 in glioma stem cells,respectively(t=3.304,P=0.01).The relative m RNA level of CD133 was 1.09±0.12 in U87 glioma cel I line, 1.80±0.13 in glioma stem cells,respectively(t=3.962,P=0.00).The relative expression level of AR protein was0.54±0.09 in U87 glioma cel I line, 0.97±0.09 in glioma stem cells,respectively(t=3.45,P=0.00)). The relative expression level of CD133 protein was0.34±0.03 in U87 glioma cel I line, 0.99±0.08 in glioma stem cells,respectively(t=7.45,P=0.00).(2) The independent risk faetors related to the glioma included family history of cancer(OR=3.270,P=0.003 and high salt diet(OR=2.774,P=0.011).Conclusion(1) The co-expression of AR/CD133 may suggested us that AR and CD133 may play an importent role in the occurrence and development of gliomas.Targeted therapy point to AR/CD133 may obtain very good curative effect(2) The family history of cancer may contacted with the occurrence and development of gliomas. The community health service could play an importent role in treatment and prevention of glioma.
