Objective: Through the use of low molecular weight heparin intervention in human lung adenocarcinoma cell line A549, detecting factors on the growth of VEGF, TGF-β1 affect the expression. Discussing low molecular weight heparin on human lung adenocarcinoma cell line A549 whether inhibitory effects and its possible mechanism. Methods:(1) The in vitro cultured human lung adenocarcinoma A549 cells in logarithmic phase is divided into not to join the low molecular heparin group(control group), low molecular heparin intervention group(concentrations were 0.5 ug/ml, 1.0 ug/ml, 2.0 ug/ml, 5.0 ug/ml, 10.0 ug/ml), a total of 6 groups, with MTT method groups was detected in 24 hours, 48 hours, 72 hours of absorbance values, and calculate the groups of proliferation inhibition rate, the data between control group and experimental group.(2) The selection of low molecular heparin best inhibition of human lung adenocarcinoma A549 cells time 72 h, extract and centrifugal supernatant on cell at the time, method of enzyme linked immunosorbent(ELISA) to detect each group on human lung adenocarcinoma A549 cells supernatant, the content of VEGF and TGF-β1 in between each group of VEGF, TGF-β1 comparison, and VEGF, TGF-β1 correlation analysis. Results :(1) Low molecular heparin intervention 24 h IOD value was 0.719±0.033 in control group, each group IOD values were 0.671±0.041, 0.632±0.049, 0.604±0.017, 0.574±0.027, 0.559±0.035; Cell inhibition rate were 6.67%, 12.10%, 15.99%,20.17%, 22.25%, the control group there was no statistically significant difference compared with the experimental group IOD values(P>0.05).(2) Low molecular heparin intervention 48 h IOD value was 0.889±0.065 in control group, each group IOD values were 0.711±0.044, 0.630±0.057, 0.586±0.091, 0.514±0.039, 0.471±0.024; Cell inhibition rate were 20.02%, 29.13%, 34.08%, 42.18%, 47.02%, control group and experimental group and the experimental way between IOD values comparative difference was statistically significant(P<0.05).(3) Low molecular heparin intervention in 72 h IOD value was 1.104±0.092 in control group, each group IOD values were 0.816± 0.027, 0.708±0.042, 0.635±0.028, 0.542±0.071, 0.450±0.036; Cell inhibition rate were 26.09%, 35.87%, 42,48%, 50.91%, 59.23%, control group and experimental group and the experimental way between IOD values comparative difference statistically significant(P<0.05).(4) Enzyme linked immunosorbent(ELISA) to detect 72 h groups adenocarcinoma A549 cell supernatant on the content of VEGF and TGF-β1, the results show that the experimental group compared with control group A549 cell VEGF and a decrease in the expression of TGF-β1, and the difference is statistically significant(P<0.05). Calculation of VEGF and TGF-β1 correlation coefficient is 0.934, which is significant positive correlation(P<0.05).Conclusion:(1) Low molecular weight heparin can inhibit human lung adenocarcinoma A549 cell proliferation, and the inhibition rate increased with time extending, in a time-dependent manner.(2) The mechanism of low molecular weight heparin inhibition of lung adenocarcinoma A549 cells may be associated with the down-regulation of expression of VEGF in human lung cancer cells and TGF-β1.(3) VEGF and TGF-β1 to play synergetic action in lung cancer development.
