Objective:By screening the anti-inflammatory of Linoleic acid,Ostrich oil,Linoleic acid methylester,Emu oil and purple sweet potato anthocyanins early,make sure that purple sweet potato anthocyanins is the main research object.The study is designed to investigate the effect of purple sweet potato anthocyanins on chronic,acute alcoholic liver injury and SNU-387 cells with underlying mechanisms.Methods:Preparation of model mice,intragastric administrated with purple sweet potato anthocyanins,investigate the effect of protection by General observation,Organ index determination,biochemical indicator (ALT,AST,ADH,GRAC) detection and HE staining pathological analysis.Tackled with different concentrations of purple sweet potato anthocyanins, activity determination and morphology observation of human hepatocellular carcinoma cells SNU-387 was made by using the MTT and the Hoechst 33258 staining.The TNF-a content was measured to investigate the effects of extrinsic apoptotic pathway medisted by TNFR1 death receptor.The changes of Caspase-8 was measured to investigate the effects of NF-κB and MAPK signal pathway by inhibitor application of PDTC and SB203580.Tackled with different concentrations of purple sweet potato anthocyanins,the reactive oxygen species content of human hepatocellular carcinoma cells SNU-387 was measured by using H2DCFDA staining with inverted phase contrast microscope and biochemical multifunctional microplate reader.At that time,the SOD activity were determined.The expression changes of Caspase-8 was measured to investigate the effects of reactive oxygen species on signal pathway by N-acetylcysteine and signal pathway blocker application.Results:In experiment of chronic alcoholic liver injury,the model group had lift on AST,ALT activity and liver index,spleen index,while a lower in ADH content and kidney index.However,anthocyanin dose group had a lower in AST,ALT activity and liver,spleen index,while a lift in ADH content and kidney index.And the HE staining pathological section showed that cells were swelled and structure was under recovery.At that time,nutritional status of riboflavin was’t impacted.In the experiment of acute alcoholic liver injury,the model group had a lift in AST,ALT activity,while a lower in ADH content.What’s more,anthocyanin dose group had a lower in AST,ALT activity and a lift in ADH content,but the range was lower.In the experiment of cell apoptosis,results showed that the activity of cells SNU-387 reduced, features of apoptotic appeared,TNF-α content and Caspase-8 expression increased. And Caspase-8 expression was no difference with PDTC,while had a lower with SB203580.In experiment of ROS,the results showed that the content of ROS increased and got SOD activity lower.The expression of caspase-8 had a lower by N-acetylcysteine application.And Caspase-8 expression was no difference with PDTC,while had a lower with SB203580.Conclusion:Purple sweet potato anthocyanins could reduce the activity of AST and ALT,increase the content of ADH,swell cells and maintain cells structure,not affect riboflavin.The protection effect on alcohol withdrawal was better than under continuous drinking.Purple sweet potato anthocyanins could participate in the apoptosis of SNU-387 cells by apoptotic pathway medisted by TNFR1 death receptor and MAPK signal pathway,improve content of ROS by limiting activity of SOD.Reactive oxygen species might active NF-κB signal pathway in a certain extent.,and could active MAPK signal pathway to participate in and promote the apoptosis of SNU-387 cells...
