Background:Schistosomiasis is a tropical disease that proposed by United Nations Development Programme,The World Bank and WTO. Schistosomiasis remains a major parasitic disease, with 200 million people infected and 779 million people at risk worldwide.Especially in Africa,South America and Asian. In the beginning of the infection, the host presents advantages of Thl response, subsequent Th2 immune deviation. The mechanism has not been clear. This study through express and purified SjFBPA to stimulating cells and in vitro studies SjFBPA induced Th2 immune response mechanism. To reveal the insect body immune escape, host immune tolerance the molecular mechanism of the immune deviation of research has important significance. Objective To analyze the dynamics of IL-4、CD8a in spleen cells of mice infected with cercariae of Schistosoma japonicum or SjFBPA and investigate the relationship between SjFBPA and the Th2-dominant response. Methods Ten mice were infected by cercariae and 10 mice were uninfected.8 weeks after infection, all mice were sacrificed. Spleen single cell suspension was prepared and flowed cytometry. Analysis was performed on the IFN-r,IL-4,CD8a cells of spleen cells stained with anti FITC-CD4, PE-IFN-r, PE-CD8a, APC-IL-4 monoclonal antibody. Results At 8 weeks, the percentage of IL-4 and CD8a cells in infected with cercariae of Schistosoma japonicum mice was24.5% and 7.65%, respectively while the percentage of IL-4 and CD8a cells in uninfected mice wasl.18% and 0.571%, indicating a gradual decline (P<0.05). The percentage ofIL-4 and CD8a cells in infected with SjFBPA was18.3% and 5.82%, the percentage of IL-4 and CD8a cells in uninfected mice was0.91% and 0.086%, indicating a gradual decline (P<0.05).Conclusion This study suggested that SjFBPA may have a potential value for the Th2 response. DC cells as presenting cells involved in the immune response.
