Objective:to study the therapeutical effect of chrysin and hydroxyethyl chrysin on MCAO(middle cerebral artery occlusion) and CCH(chronic cerebral hypoperfusion); and then to study their mechanisms.Method: operating middle cerebral artery occlusion surgery or improved permanent bilateral common carotid artery occlusion surgery to create animal model, using nervous function score,8-Arm Maze,TTC-staining, Nissl’s staining and HE-staining to evaluate the grade of damage, western blotting and biochemistry indicators are used to explore its mechanisms;Result:compared with the sham operation group, the cell structure of MCAO group is damaged according to HE-staining and Nissl’s staining, nerve function score and infarction volume remarkably increased, T-AOC and CAT activity, GSH-PX activity, SOD activity declined, the expression of Nrf2, HO-1, NQO1, GCLC, GCLM and apoptosis related Bcl-2, Bax upregulated; in Chrysin group and HC group, compared with the MCAO group, the damage induced by ischemia, nerve function score and infarction volume decreased, T-AOC and CAT activity, GSH-PX activity, SOD activity increased, the expression of Nrf2, HO-1, NQO1, GCLC, GCLM and Bcl-2 significantly upregulated.Compared with the sham operation group, the cell structure of CCH group is damaged according to HE-staining, the cognitive ability declined, CAT activity and GSH-PX activity declined, T-AOC and SOD activity increased, the expression ofNrf2, HO-1, NQO1, GCLC, GCLM and apoptosis related Bcl-2, Bax upregulated; in Chrysin group and HC group, compared with the CCH group, the damage induced by ischemia decreased, the cognitive ability improved, CAT activity and GSH-PX activity increased, T-AOC and SOD activity decreased, the expression ofNrf2, HO-1, NQO1, GCLC, GCLM and Bcl-2 significantly upregulated.Conclusion: the therapeutical effect of chrysin and HC on MCAO and CCH is related to upregulate Nrf2-ARE pathway.
