Expression of Shh, Gli1, Cyr61 in Breast Cancer and Pathological SignificanceObjectiveThe purpose of this study was to detect the expression of Shh, Gli1 and Cyr61 protein in breast tissue and investigate the association among Shh, Glil, Cyr61 and pathological factor respectively and analyze the correlation between Cyr61 and Shh signaling pathway.Methods124 breast invasive ductal carcinoma cases,15 breast cancer adjacent tissue,17 mastoplasia tissue,22 breast fibroadenoma tissue and their relevant clinical data were collected from the Pathology department, Hospital of Hexi Universuty, Gansu province. Streptavidin-perosidase (SP) was used to detect Shh, Glil and Cyr61 in breast tissue.Results1. Immunohistological postive expression rate of Shh in breast cancer adjacent tissue, benign breast lesions and breast invasive ductal carcinoma tissue were 13.3%, 43.6% and 49.2%, respectively, and the result was significantly different(P<0.05); the positive rate of Glil in breast cancer adjacent tissue, benign breast lesions and breast invasive ductal carcinoma tissue were 20.0%,56.4% and 38.7%, respectively, which showed a significant difference (P<0.05); The positive rate of Cyr61 in breast cancer adjacent tissue, benign breast lesions and breast invasive ductal carcinoma tissue were 26.7%,48.7% and 69.4%, respectively, which demonstrated a significant difference (P<0.05).2. Shh protein expression was significantly associated with the tumor size and positive axillary node status in breast cancer patients (P<0.05).3. There was a significant positive correlation between the expression of Shh and the level of GST-π (rs=0.196, P<0.05). Cyr61 was positively correlated with the expression of HER-2 (rs=0.223, P<0.05) and P53 (rs=0.330, P<0.01).4. In breast cancer, Shh protein expression was in positive correlation with the expression of Gli-1 (rs=0.311, P<0.01) and Cyr61 (rs=0.234 P<0.01). Gli-1 protein expression was positively correlated with Cyr61 (rs=0.313, P<0.01).5. Univariate survival analysis of the correlation with lymph node metastasis, pTNM stage and overall survival was significantly difference in breast cancer (P< 0.05).Conclusion1. High expression of Shh, Gli-1 and Cyr61 in breast cancers suggested that Shh signaling pathway was activited in breast cancer and these indexes may be used as biomarkers for the diagnosis of breast cancer.2. Shh protein expression correlated with the tumor size and positive axillary node status suggested that Shh may promoted the growth of tumor, which is associated with poor prognosis and may be used as a index of prognosis.3. There was a significant positive correlation between the expression of Shh and the expression level of GST-π. Cyr61 was positively correlated with the expression of GST-π and P53, which suggested that Shh may be relate to the resistance in breast cancer cell and the prognosis of patients.4. Shh protein expression was in positive correlation with the expression of Gli-1 and Cyr61. Gli-1 protein expression was positively correlated with Cyr61, suggested that Cyr61 was associated with Shh and Cyr61, and which may be an important regulatory factor for Shh signaling pathway.5. Univariate survival analysis of lymph node metastasis and pTNM stage was associated with with the overall survival of breast cancer patients, prompt lymph node metastasis and pTNM stage was associated with the prognosis of patients with breast cancer may be, but not as an independent factor affecting the prognosis of patients with breast cancer.Expression of Shh, Gli1, Cyr61 in Gastric Cancer and Pathological SignificanceObjectiveThe aim of this study was to detected expression level of Shh signaling pathway related protein such as Shh, Glil and Cyr61 in gastric cancer patients, analyze its relationship in clinicopathological parameters among the Shh, Glil and Cyr61 protein, investigate the relationship between Cyr61 and Shh signaling pathway, and prove the feasibility of the Shh signaling pathway and Cyr61 as a new target in gastric cancer.Methods15 chronic gastritis tissue cases,49 gastric adenocarcinoma case,24 gastric mucinous adenocarcinoma case,17 gastric mucous cell carcinoma case and their relevant clinical data were collected from the Pathology department, Hospital of Hexi Universuty, Gansu province. Streptavidin-perosidase (SP) is used to detect Shh, Glil and Cyr61 in gastric cancer.Results1. Shh expression rate in chronic gastritis tissue and gastric carcinoma were 20.0% and 71.1%, respectively, Glil expression rate in chronic gastritis tissue and gastric carcinoma were 13.3% and 65.6%, respectively, Cyr61 expression rate in chronic gastritis tissue and gastric carcinoma were 33.3% and 78.9%, respectively, All the results were statistically significant difference (P<0.05, respectively).2. There was some relevance among Shh expression, tumor invasion, lymph node metastasis and pTNM stage in gastric cancer, the difference was statistically significant. (P<0.05); However, the Cyr61 expression and tumor invasion was not significantly different in gastric cancer (P>0.05)。3. There was a significant positive correlation between the expression of Shh and the expression level of Ki-67 (rs=0.337, P<0.01), Cyr61 was positively correlated with the expression of GST-π (rs=0.355, P<0.01), Cyr61 was positively correlated with the expression of Ki-67 (rs=0.539, P<0.01)4. In gastric cancer, Shh protein expression was in positively correlated with the expression of Gli-1 (rs=0.621, P<0.01) and Cyr61 (rs=0.631, P<0.01), Gli1 protein expression was positively correlated with Cyr61 (rs=0.427, P<0.01)5. The single factor analysis showed the overall survival was not significantly different in the high/low expression of Cyr61, Shh, Gli1 groups in gastric cancer patients (P> 0.05). Tumor infiltration depth and pTNM stage was significantly related with overall survival (P<0.05); Multivariate analysis showed TNM stage was obviously related to the prognosis of gastric cancer patients (P> 0.05).Conclusions:1. High expression of Shh, Gli-1, and Cyr61 in gastric cancers, suggested that Shh signaling pathway was activited in gastric cancers and the these indexes can be used as biomarkers for the diagnosis of gastric cancer patients.2. There was some relevance among the Shh expressed in gastric cancer and the tumor invasion, lymph node metastasis and pTNM stage, and there was a relevance between the Cyr61 expressed in gastric cancer and the tumor invasion, by explaining Shh and Cyr61 can promote invasion and metastasis of gastric cancer cells, which is associated with poor prognosis and can be used for index of prognosis.3. There was a significantly positive correlation between the expression of Cyr61 and the expression level of GST-π. Shh positively correlated with the expression of Ki-67, Cyr61 positively correlated with the expression of Ki-67, tips Cyr61 may be related to the resistance of gastric cancer cell, Shh and Cyr61 are concerned with the proliferation of gastric cancer cells, which may be increased proliferation of gastric cancer cells and promote the development and the prognosis of gastric cancer patients.4. There was a significantly positive correlation between the expression of Shh and the expression level of Gli-1, Shh positively correlated with the expression of Cyr61, Gli-1 positively correlated with the expression of Cyr61, tips Cyr61 may be related to the Shh signaling pathway, Cyr61 may be role as an important regulatory factor in Shh signaling pathway.5. Multivariate analysis of pTNM staging and correlated with overall survival in patients with gastric cancer, prompt pTNM staging is associated with the prognosis of patients with breast cancer may be, can be used as the independent factors affecting the prognosis of patients with breast cancer.
