Objective:With the enlargement of the application of nuclear technology, especially the emergence of the nuclear power plant and radiation processing technology. Radiation accidents have increase trend, caused serious environmental radioactive pollution, personal injury and economic losses.In order to find a high efficiency and low toxicity radiation protector, in this paper, it studied the radiaition protective effect of pyrrolidine dithiocarbamate (PDTC) and made the structure modification on PDTC. Provide experimental basis for looking for new radiation protection agent.Methods:1. ICR mice were selected as investigated subject, then the mice were whole body irradiated by 7.5Gy or 4.5Gy?5.0Gy/6.0Gy137Cs y-ray. The 30-day survival rates, organ indexes, peripheral blood and hematopoietic function and T-SOD> MDA served to evaluate the radiation protective effect of PDTC.2. ICR mice were selected as investigated subject, then the mice were whole body irradiated by 6.0Gy 137Cs y-ray. The organ indexes, peripheral blood and hematopoietic function observed to evaluate the combined radiation protective effect of PDTC and Amifostine.3. PDTC as the lead compound was combinated with 5- methoxyl primary amine, isoamyl alkenyl, allyl and got three compounds:Compound A、B、 C. ICR mice were selected as investigated subject, then the mice were whole body irradiated by 6.0Gy137Cs y-ray. The organ indexes, peripheral blood, DNA content of marrow and colony forming unit of spleen (CFU-S)observed to evaluate the radiation protective effect of compound A.Result:Pharmacological experiment results showed that PDTC increased the 30-day survival rates of 7.5Gy whole body irradiated mice, the 60 mg/kg dose group was the most obvious. And after 4.5、5.0、6.0Gy of whole body irradiation. Under the 4.5 or 5.0Gy irradiation dose,the effect of 30 mg/kg is relatively good, but under 6.0Gy dose,60 mg/kg and 90 mg/kg group showed good effect. Compound A could enhance the organ indexes、peripheral blood indexes, colony forming unit of spleen. It had certain radiation protection in radiation injury mice. Three compounds were synthesized with PDTC as the lead compound.The chemical structures were confirmed by MS and 1HNMR.Conclusion:PDTC has certain radiation protection function. Three compounds were synthesized with PDTC as the lead compound and if s confirmed the salt derivatives has certain radiation protection.
