Background and ObjectiveHypopharyngeal squamous cell carcinoma (HSCC) is a common malignant tumor in head and neck, invasion and metastasis are responsible for the high death-rate. It associated with smoking, drinking, along with the add greatly to air pollution, the morbidity of HSCC is high recent years. The treatment of many HSCC patients was delayed before because of the depth of lesion location, the slur of early symptom and the lateness of correct diagnosis. The vast majority of them were clinical III, IV period of advanced carcinoma, Laryngeal cavity, cervical esophageal, and the root of tongue were often involved, and most with cervical lymph nody metastasis. After surgery may lead to pronunciation, breathing and swallowing dysfunction, and influence the patient’s quality of life seriously. Recurrence, cervical lymph nody metastasis and distant metastasis were the main factors influencing the prognosis of patients and lead to the failure of treatment. A better understanding of the pathogenesis of HSCC and identification of its molecular alterations are essential for the development of novel effective therapies for HSCC. Therefore, studies of novel and more effective molecular biomarkers of HSCC prognosis and progression are still necessary, Its helpful to the early diagnosis, early treatment and improve prognosis of HSCC patients.With the rapidly development of functional genomics, more and more attention focus on the study of non-coding transcription product functions. Non-coding RNA is a kind of RNA molecules which do not have coding protein function, but they play important and extensive regulation functions in the life activities. Relative to the protein coding sequence and all kinds of small RNA, long non-coding RNA (lncRNA) is a hotspot in the current research on non-coding RNA. Many studies have shown that lncRNA plays a variety of different functions in the process of cancer occurrence and development (promoting cancer effects or anticancer effects). They are involved in the tumor cells apoptosis regulation, invasion and metastasis. They had hoped to become a new tumor markers and even the target of tumor therapy, and showed good prospect of clinical application in cancer diagnosis, treatment and evaluation of the prognosis of patients. In the present study, we aimed to find new lncRNAs that correlate with HSCC and test whether the expression of those lncRNAs could be a potential biomarker for HSCC.MethodsWe chosen three paired primary cancerous and adjacent noncancerous tissue samples were used for global profiling of human lncRNA expression using the Arraystar Human lncRNA Microarray (Arraystar, Rockville, MD, USA). We found several differentially expressed lncRNAs and protein-coding genes in in hypopharyngeal squamous cell carcinoma (HSCC),there were ABO 19562,AB209630, SPP1,TJP2, respectively. Additionally, twenty HSCC specimens and their matched noncancerous mucosal epithelial tissues were obtained for confirmation of differential lncRNA expression by qRT-PCR.ResultsThe expression of AB209630 was down regulated in HSCC tissue.The effect of AB209630 overexpression on the colony formation of FaDu cells was analyzed by colony formation assay, The results of MTT experiment and Transwell experiment showed that:the over-expression of lncRNA-AB209630 could inhibit the proliferation and invasion of HSCC cell line Fadu, suggesting this AB209630 was a potential tumor suppressive lncRNA in HSCC. Importantly, head and neck cancer cell lines with lower level of AB209630 expression are more resistant to irradiation treatment.ConclusionThe clinical significance and biological function of lncRNAs in hypopharyngeal squamous cell carcinoma (HSCC) remain largely unknown. We characterized the novel lncRNA AB209630 in vivo and in vitro.The preliminary studies demonstrate the LncRNA-AB209630 in proliferation..invasion and metastasis of HSCC play an important regulatory role in the process, these results provided new experimental evidences and research ideas for the mechanism, development, invasion and metastasis and prognosis of HSCC, the results from this project will lead to better understanding of the molecular mechanisms underlying the carcinogenesis of HSCC, and could identify novel therapeutic targets for the treatment of HSCC.
