BackgroundColorectal cancer, one of the most common malignant tumor that is threat to human health seriously. Its incidence in men and women is the third most common malignant tumor worldwide, and the fourth cause of cancer mortality. In China, according to incomplete statistics, the incidence and mortality of colorectal cancer in malignant tumor is the fourth and fifth respectively, and it has a tendency to rise and to younger in recent years. The pathogenesis of colorectal cancer is not clear. It is believed to be a multistep process that involves diet, environment, genetic and physical activity, in addition to multiple genetic changes. Recent studies have shown that CCRK (cell cycle related kinase) can promote the cell cycle progression and positive regulation of cell cycle process. Some studies have found that CCRK is overexpressed in many malignant tumors. CCRK knockdown will causing cell cycle arrest and decreased cell proliferation. CCRK involved in signaling pathways associated with the tumor formation and evolution. It has not yet been reported that whether CCRK gene is participate in the occurrence of colorectal cancer and the relationship with malignant biological characteristics. Next, we’ll study CCRK expression in colorectal cancer and the relationship between clinical pathological features.AbjectiveTo compare the CCRK expression levels in colorectal cancer and adjacent colon tissue, and in different cases of patients with colorectal cancer, in order to clarify CCRK expression with colorectal cancer occurrence and its relationship with tumor pathology classification and transfer, to explore the early molecular biology mark in colorectal cancer occurrence and stages, providing the basis for the early diagnosis, prognosis and treatment of colorectal cancer.Methords1. RT-PCR was performed to examine the CCRK mRNA expression in 50 colorectal cancer tissues and adjacent normal colon tissues.2. Western-blot was performed to detect the CCRK protein expression in 50 colorectal cancer tissues and adjacent normal colon tissues.3. Immunohistochemistry staining (IHC) study was performed to detect CCRK expressed in different stages colorectal cancer.4. Analized the relationship between CCRK expression and clinicopathological features, such as gender、age、tumor size、location tumor grade and clinical stages.5. Statistical analysis was performed with SPSS 17.0 for all datas. The value of CCRK mRNA expression and protein expression were assessed by y±S two groups. The comparison of two groups was assessed by t-test. Chi-square test was used to assess the expression rate in two groups. The differences were considered to be statistically significant at a value of P less than 0.05.Results1. RT-PCR results:The positive expression rate of CCRK mRNA in colorectal cancer tissue 64%(32/50) was significantly higher than in adjacent colon tissue 42% (21/50, x2= 4.857, p<0.05).The CCRK mRNA relative expression level in colorectal cancer tissue (2.45±0.92) was significantly higher than that in adjacent colon tissue (1.00±0.21) (t=4.83,P<0.05).2. Western-blot results:The positive expression rate of CCRK protein in colorectal cancer tissue 60%(30/50) was significantly higher than that in adjacent colon tissue 40%(20/50).The difference was statistically significant in two groups (X2=5.769, p<0.05).The CCRK protein relative expression level in colorectal cancer tissue (2.69±0.87) was significantly higher than that in adjacent colon tissue (1.00±0.28), and the differences were statistically significant (t=5.81,P<0.05). The expression of CCRK protein was weak in adjacent carcinoma tissue. The positive expression of CCRK protein in colorectal cancer tissue and in adjacent carcinoma tissue that was in consistent with CCRK mRNA expression.3. Immunohistochemistry staining results:CCRK protein positive expression in colorectal cancer tissue mainly located in cytoplasm and nuclear membrane, a small amount in the nucleus. The positive expression rate of CCRK in colorectal cancer tissues was 52.7%(79/150),significantly higher than the adjacent tissue section of 35.2%(58/165), there is statistical difference between two groups (χ2= 9.81, p< 0.05).4. CCRK expression and clinical pathological features:It showed that a significant positive association between CCRK ectopic expression and patients clinical stage (P=0.046).This study showed that CCRK mRNA positive expression rate of the PT3-4 stage colorectal cancer tissue was 85.7%(12/14) which was significantly higher than that the PT1-2 stage of 55.6(20/36,χ2=3.979,p=0.046). CCRK mRNA positive expression rate in PN1-2 stage colorectal cancer tissues 87.5% (14/16) was significantly higher than in PNO stage of 52.9%(18/34,χ2= 5.640, p=0.018). With the tumor progress, the expression of CCRK increases gradually. No significant association with the patients gender、age、tumor size and location as well as tumor grade and PM status(P>0.05).Conclusion1. CCRK high expression associated with the incidence of colorectal cancer. The expression quantity is higher, the greater the probability of colorectal cancer incidence. And its expression is related to its clinical classification, the higher degree of differentiation, the expression quantity is lower. It indicated that CCRK played a promoting role in the occurrence and development of colorectal cancer.2. CCRK may become a new reference indicator in colorectal cancer diagnosis and prognosis.
