ObjectiveTo study the expressions and Clinical significance of hypoxia inducible factor-la (HIF-la), vascular endothelial growth factor (VEGF), vascular endothelial g rowth factor receptor-1 (VEGFR-1) and vascular endothelial growth factor recept or-2 (VEGFR-2) in the villi tissue of patients with missed abortion (MA) and early pregnant women,explore the influence of hypoxia on placenta angiogenesis in MA.Methods35 patients of missed abortion were collected and categorized into research proup from Jun 2013 to Dec 2013,they were all diagonised by B-ultrasound and P-HCG continous monitoring;meanwhile,32 patients of early pregnancy with artificial surgery were categorized into control group,which were proved to be intrauterine pregnancy/visible embryonic heart beats tube by B-ultrasound.To detect the expression of HIF-1α、VEGF、 VEGFR-1、VEGFR-2 in the Villi tissue of research group and control group by Immunohistochemistry,and the results were statistical analysis.Results1 Expression of HIF-1α in villous:the expression of HIF-la were expressed both in research group and control group,mainly expressed in the cytoplasm and nucleus of cytotrophoblastic> syncytiotrophoblastic、cytoplasm of endothelial cells surrounding stroma cells.,which was higher in research group than in control group (P<0.05)2 Expression of VEGF in villous:the expression of VEGF were expressed both in research group and control group,mainly expressed in the cytoplasm of cytotrophoblastic、syncytiotrophoblastic、endothelial cells surrounding stroma cells.,which was lower in research group than in control group (P<0.05)3 Expression of VEGFR-1 in villous:the expression of VEGFR-1 were expressed both in research group and control group,mainly expressed in the cytoplasm of cytotrophoblastic、syncytiotrophoblastic,which was slightly higher in research group than in control group.4 Expression of VEGFR-2 in villous:the expression of VEGFR-2 were expressed both in research group and control group,mainly expressed in the cytoplasm of cytotrophoblastic、syncytiotrophoblastic、endothelial cells surrounding stroma cells.,which was significantly lower in research group than in control group (P<0.01).5 The correlation analysis of HIF-la and VEGF、VEGFR-1、VEGFR-25.1 In research group:There is correlation between the expression of HIF-la and VEGF、VEGFR-2.(DThe Pearson colleration coefficient of HIF-la and VEGF is r=-0.334, P=0.043<0.05,showed a negative correlation between the expression.②T he Pearson colleration coefficient of HIF-1α与VEGFR-2 is r=-0.406, P=0.015<0. 05,showed a negative correlation between the expression.The Pearson colleration coefficient of HIF-la与VEGFR-1 is r=0.162, P=0.352>0.05,showed no correlati on between the expression.5.2 In control group:There is correlation between the expression of HIF-la and VEGF、 VEGFR-1、VEGFR-2.①The Pearson colleration coefficient of HIF-la and VEGF is r=0.534, P=0.002<0.01,showed a significantly positive correlation between the expression;(2)The Pearson colleration coefficient of HIF-la与VEGFR-1 is r=0.381, P=0.031<0.05,showed a positive correlation between the expression;③The Pearson colleration coefficient of HIF-1α与VEGFR-2 is r=0.402, P=0.023<0.05,showed a positive correlation between the expression.Conclusions1 Expression of HIF-la in villous of MA is higher than induced abortion.Suggset the increased HIF-la is associated with MA.2 Expression of both VEGF and VEGFR-2 in villous of MA are higher than that of induced abortion. Suggset the decreased of VEGF and VEGFR-2 are associated with MA,which induced the decrease of angiogenesis of the placental villi which nourish inadequate blood supply to trophoblasts,causing embryonic development cease.This may be one of the important reasons for MA.3 Expression of HIF-1α has negative correlation with both VEGF and VEGFR-2 In MA. Suggest HIF-la inhibit VEGF and VEGFR-2 genes transcription in severe hypoxia, the imbalance of the relationship between HIF-la and VEGF、VEGFR-1、 VEGFR-2 may give rise to the abnormal placental vascular development,which may be the important cause of MA and closely related with MA.
