| Objective: To establish the mode of the middle cerebral artery occlusion(MCAO) of rat model through reversible suture method, and explore the expression of p-38 MAPK NF-κB and MMP-9 on cerebral ischemia-reperfusion injury, andto analyze the influence of diammonium glycyrrhizinate. Methods Forty-Two male SD rats were randomly divided into the normal group(group A, 6 rats), the diammonium glycyrrhizinate group(group B, 15 rats), the saline group(group C, 15 rats) and the model control group(group D, 6 rats). The rat in group B, C and D were established MCAO model through reversible suture method. The rat model in group B were injected with diammonium glycyrrhizinate, and the rats in group C were injected with saline group. The neurological was assessed by Zea-Longa score. All rats were sacrificed after 3 days, the cerebral infarction area was detected by TTC staining, and the expression of p-38 MAPK was detected by Western blot,Immunohistochemical staining detecting Bcl-2,NF-κB and MMP-9 protein expression.Results: The Zea-Longa scores were respectively(3.60±0.51) scores,(3.53±0.52)scores, and(3.67±0.52)scores in group B, C and Dafter modeling, and MCAO model wasestablishing.The cerebral infarction area and p-38 MAPK expression in group A were significantly less than the group D(p<0.05), and the Zea-Longa scores, cerebral infarction area and p-38 MAPK expression in group B were also significantly less than the group C(p <0.05). But the above indicators were showed no significant difference between group C and D(p >0.05);NF-κB immunohistochemical staining: A group(8.16±1.16), group B(30.00±2.60), group C(42.50±3.78), D group(40.89±2.04), group B and C, D between the two groups there was significant difference(p<0.01), whereas no significant difference(P between groups C and D> 0.05); MMP-9 immunohistochemical staining results: A group(9.00±2.09), group B(14.83±1.16), group C(19.00±2.36), D group(17.33±2.50),group B and C,D are more differences between the two groups was statistically significant(P <0.05), and between groups C and D groups was not statistically difference(P = 0.18). Bcl-2 immunohistochemical staining and apoptosis Tunel comparison: Bcl-2 immunohistochemical staining and apoptosis were Tunel A group(49.83±4.40),(7.16±1.16), group B(52.33±3.07),(8.17±1.47), group C(6.66±1.21),(51.83±4.40), D group,(10.16±1.47),(50.00±3.09). Group B and C, D groups are more statistically significant differences(p <0.01), whereas no significant difference(P between groups C and D> 0.1), and no significant difference between group A and group B(p > 0.05).Conclusions: Licorice diammonium significantly reduced ischemia-reperfusion injury of infarct size and neurological score, and can reduce the expression of p-P38 MAPK inflammatory pathways and NF-κB, and reduces MMP-9 expression of inflammatory cytokines,upregulate apoptosis-related protein Bcl-2 expression and reduce the Tunel staining, thereby alleviating ischemic reperfusion injury. |
