| Background and Objective:Male infertility becomes a more and more serious social issue, it was reported that male factors account for fifty percent in those infertile couples.Azoospermia patients are faced with more challenges.In many cases,patients who have endocrine or obstructive diseases can be treated medically or surgically,but those who suffer idiopathic azoospermia or testicular failure have less hope.Up to now,we can only pin our hope on the translation of stem cells.In this experiment,Two methods of injecting busulfan to SD rats are taken to establish a model of “Spermatogenesis dysfunctionâ€and lay the first stone for the model of “Sertoli cell only syndromeâ€. Methods:140 SD rats were divided into two groups which has 70 rats each,intraperitoneal injection of Busulfan and testicular injection of Busulfan were taken to attempt to establish different experimental animal models, survival rate, routine blood test and testicular pathology analysis were compared between them. Results:70 SD rats which experienced intraperitoneal injection of busulfan all died within one month, before death the rats showed dimmed iris, eyelid crusting, slow activity and so on, some rats showed bleeding around nose and anus(57.5%), Blood analysis showed that red blood cells, hemoglobin and platelets decreased significantly(P< 0.05). Johnsen score was no significant difference compared with control group(P >0.05). SD rats which experienced testicular injection of Busulfan observed no deaths within 30 days, Blood analysis revealed that red blood cells, hemoglobin and platelets compared with control group showed no significant sifference(P>0.05). Johnsen score was significantly difference compared with control group(P<0.05). Conclusion:Intraperitoneal injection of busulfan may cause serious death in rats.Testicular busulfan injection in rats showed no significant toxic effects, by 0.1mg/Kg,0.2 mg/Kg,0.3 mg/Kg,0.4 mg/Kg,it can be successfully uesd to establish the model of“Spermatogenesis dysfunctionâ€,and there may be a suitable dosage for “Sertoli cell only syndrome†model,so it is a new method and worth of further research. |
