Objective To study the Long chain of non-coding RNA HOTAIR expression a nd its role in CD133 positive gastric cancer cells.Methods Magnetic cell sorting,MACS sorting CD133 positive and CD 133 negat ive cells in KATO-Ⅲ gastric cancer cells. Reverse transcription polymerase chain reaction (RT-PCR) detect the HOTAIR and CD133 mRNA expression difference s. Small RNA interference technology (Small interfering RNA, siRNA) interferen ce the expression of HOTAIR in CD 133 positive KATO-Ⅲ cells, RT-PCR detect the efficiency of RNA interference and detect the impact of HOTAIR interferen ce on the mRNA expression of CD133 and EMT related factor. Transwell detect the influence of HOTAIR interference on cell migration and invasion ability.Results After MACS separation, the CD 133 mRNA expression in the group of CD133 positive cells(0.6056±0.0163) were significantly higher than those of CD1 33 negative group(0.4337±0.0115,p=0.000) and no separation group(0.4451±0.0175, p=0.000).After MACS separation, the HOTAIR mRNA expression in the group o f CD 133 positive cells(0.7143±0.0480) were significantly higher than those of C D133 negative group(0.4978±0.0133,p=0.000) and no separation group(0.6169±0.00 84,p=0.006). After interference the HOTAIR RNA expression on CD133 positive KATO-Ⅲ cells, the expression of HOTAIR mRNA in siH group(siH)(0.3555±0. 0110) was lower than that in group NC(NC)(0.6326±0.0212, p=0.000) and contro 1 group(Ctrl)(0.6620±0.0162, p=0.000), the expression of CD133 mRNA in siH g roup(siH)(0.4396±0.0181) was lower than that in group NC(NC)(0.6221±0.0145,p= 0.000) and control group(Ctrl)(0.6342±0.0089, p=0.000), the expression of E-cadh erin mRNA in siH group(siH)(0.8262±0.0044) was higher than that in group NC (NC)(0.6377±0.0160,p=0.000) and control group(Ctrl)(0.6281±0.0076, p=0.000), the expression of N-cadherin mRNA in siH group(siH)(0.3426±0.0181) was lower th an that in group NC(NC)(0.6394±0.0070,p=0.000) and control group(Ctrl)(0.6287± 0.0167,p=0,000). Transwell migration experiment found that RNA interference the expression of HOTAIR in CD 133 positive KATO-Ⅲ cells, cells that through th e cell membrane in siH group(siH)(26.6667±2.5167) were significantly lower than that of NC group(NC)(68±15.6204,p=0.003) and control group(Ctrl)(69±10.1489,p =0.003). Transwell invasion experiment found that after interference the expressio n of HOTAIR in CD133 positive KATO-Ⅲ cells, cells that through the cell me mbrane in siH group(siH)(16.6667±3.2146) were significantly lower than that of NC group(NC)(36.6667±1.5275,p=0.002) and control group(Ctrl)(37.0000±7.0000,p =0.002).Conclusion The expression of HOTAIR in CD 133 positive cells was higher th an CD133 negative cells in KATO-Ⅲ gastric cancer cells, interfere HOTAIR exp ression can reduce the expression of CD133 mRNA in CD133 positive KATO-Ⅲ cells, Inhibition gastric cancer cells occur Epithelial-mesenchymal transition(E MT),inhibit gastric cancer cell migration and invasion.
