Objdvtive: Spina bifida is a clinical type of neural tube defects,the pathogenesis is not definite.With the change of living environment and conditions of huaman,the Spina bifida has become one of the main reasons affecting the neonatal mortality and disability.In the past 10 years,the study of Spina bifida in fetuses focuses on prenatal ultrasound diagnostic methods,not to pay more attention to the occurrence reason of it.In this experiment,we discus the molecular mechanisms of induced Spina bifida in fetuses through epidemiological investigation which including history collection,Ststistic-al analysis,animal model and molecular biology techniques,thereby to reduce the incidence of fetal Spina bifida and to service for human by better medicalachievement.Methods: The project analyse the statistical analysis and history collection of fetal Spina bifida incidence from Affiliated Hospital,Central Hospital,266 hospital,and maternal and child health hospital of Chengde City,Hebei Province between 2011 to 2014,then we visit 1/2 portion of patients from random sampling.During the visit,we consult the factors inducing Spina bifida detailedly,and fill out the form strictly.For the strictness and reference,we also visit the same number of normal pregnant woman which can compare the data of anteroposterior.We divide the specific items study intotwo categories: one is the genetic factors, such aspregnant who have history of birth defects or family deformitiesl; another is environment fators including the diet of pregnant women,the amount of exercise,emotion changes and the couples career.Pregnant woman in all ages and pregnance time are summarized for getting some predicting factors.Through the statistical analysis of the interview, we use the mice(C57BL/6) asexperimental model,design detection experiment.40 mice were divided into 4 groups, 10 mice in each group, three of which are treated as experimental group in different conditions, another group of normal diet as control group, regular observation using two-dimensional ultrasonic positioning technology detection of the development of mouse fetal spine under different induction conditions.Using statistical methods,we analyse the effects of three fators on fetal spine development of mouse. Firstly, whether the design is statistically significant through principal component analysis method and chi spuare qualitative method,and then using analysis of variance and correlation detection method to detect stable relationship of three factors influence on development of fetal spine.According to the research reported earlier,the expression level of nestin is concerned with proliferation and differentiation of nerve cell.We speculate that some factors may affect the expression of nestin,and the effects of different protein affects the development of fetal spine.We designe the following experiment:according to reports,the target of study is mice pregmant 10 days. 40 mice are divided into 4 groups, 10 mice in each group, three of which aretreated as experimental group in different conditions, another group of normal diet as control group.When the mice pregant 10 days,we isolate embryonic neural stem cells from fetal rat at the same time.We collecte the cell lysates, using western blot analysis to detect the expression of nestin in different groups. As β-actin a reference, using quantitative Image J software calculate the expression of nestin,forecast the three fators affecting on fetal spine bifida,and then explore the etiology of spine bifida at molecular mechanism.Results: From 2011 to 2014 between the ultrasonic detection of spina bifida(dominant) the fetus is 109, accounting for 3% of the population born, and every year there is a downward trend, followed by 38 cases in which the fetus for families to visit the work. There are eight cases which family members appeared before seriousneural tube defects, 12 cases are aged over 35 years,there are four cases which spouses or one of them in the work environment is extremely harsh chemical air coal plants work, there are 10 cases of maternal pregnanttime in summer during the prriod of high temperature,three cases of pregnant women during pregnancy have experienced a fall or collision with hard objects, there are five cases before pregnancy or taking birth control pills, there are two cases in pregnancy have alcohol consumption experience, as well as four cases without these conditions.Through the above investigation,we can realize that scientific knowledge propaganda and the government of folic acid supplements for pregnant women reduce the number of patients with spina bifida year by year.At the same time,elderly parturient women,pregnancy in high temperature and air pollution will increase the morbidity of fetal spina bifida.Conclusion: The mouse as an experimental model, by two-dimensional ultrasound positioning technology and western blot detection methods,the results of statistical analysis show that the folic acid,aging,high temperature and wicked air environment have important influence on the incidence of fetal spina bifida,and folic acid make more significant effect,the second is aging,the last is high temperature and wicked air environment.
