Objective: Astragalus Injection on prevention and treatment of contrast induced nephropathy in diabetic rats, and to provide a new way of treatment and effective for clinical medical workers.Methods: Use the SPF Wistar rats, male, weighs 200 g±20 g, model was established. Experiments are divided into normal Control group(the Control, C), model group of Diabetic nephropathy(Diabetic nephropathy, DN), Diabetic nephropathy + Contrast group(Contrast medium, CN), model + Contrast agents, astragalus injection low-dose group(low, L), model + Contrast agent + dose group, middle, M), model + Contrast agent, high dose group(high, H). Each group of five, a total of 30. Observe each group rats eating, drinking water, urine, mental state, activity, color, etc. A week time to weigh in rats. Determination of related parameters of renal; Determination of fasting blood glucose(FBG) and fasting insulin(INS) content; The determination of lipid related indicators; Observe kidney tissue pathological morphology; Determination of oxidative stress related indicators; Immune histochemical method detection in the rat kidney caspase 3 protein expression; Rt-pcr detection rat kidney the BCL- 2, bax m RNA expression.Results: Compared with normal control group, the DN set of food, drinking water and increase of urine, weight drops obviously, a swooning in spirit; Kidney index(KI), urine protein(UP), serum urea nitrogen(s BUN) and the urine urea nitrogen(u BUN), elevated serum creatinine(s Cr), urine creatinine(u Cr) discharge reduction. Contrast agents after the intervention, CN group rats kidney index(KI), urine protein(UP), serum urea nitrogen(s BUN) and the urine urea nitrogen(u BUN), serum creatinine(s Cr) further increases, urine creatinine(u Cr) excretion function decline further. And L, M, H group kidney index(KI), urine protein(UP), serum urea nitrogen(s BUN) and the urine urea nitrogen(u BUN), serum creatinine(s Cr) is reduced, and the urine creatinine(u Cr) excretion increased, and presents good dose dependent.DN group rats higher fasting blood glucose(FBG), fasting insulin(FINS), after giving astragalus injection, L, M, H group FBG decline gradually, FINS gradually rise, are dose dependent. After given astragalus injection, L, M, H group of TG, TC a dose dependent decrease; DN group microscopically glomerular injury in rats, after giving astragalus injection, L, M, H group the renal pathological changes of rats significantly reduce; DN group rats malondialdehyde(MDA) increased, the total superoxide dismutase(T SOD), catalase(CAT), glutathione peroxidase(gsh-px), after giving astragalus injection, L, M, H group presents the tendency of inhibiting oxidative stress, and a good dose dependent; After given astragalus injection, L, M, H a dose dependent decrease group of positive region, compared with the CN group have obvious difference. The Bcl- 2 DN group transcription level is reduced, and Bax transcription levels. CN group than the DN apoptosis, statistically significant(P < 0.05). After given astragalus injection, L, M, H group of apoptosis cells gradually reduce, a good dose dependent.Conclusions: Astragalus injection can improve diabetes contrast nephropathy rats model of kidney function and structural damage, and its mechanism may be largely related to its good oxidation resistance, resistance to apoptosis function.
