Objective To prepare egg yolk immunoglobulin(IgY) against P. gingivals and to study its biological characteristics in the interest of further application. Methods 150 days old laying hens were immunized with P. gingivals and IgY was isolated from the egg yolk by water dilution(WD) method and two steps of ammonium sulfate purification The titers and specificity of the antibodies were assessed by enzyme-linked immunosorbent assay(ELLSA).The molecular weight were measuredby SDS-PAGE. The antigenicity and function of the IgY were demonstrated by use of western blotting and the hemagglutination inhibition(HI) test,respectively. Results The yield of IgY was 70 mg per yolk, and the purity was 65%. Its titer was 25600.Pg-IgY binds specifically to P. gingivals and could inhibit the hemagglutination at 3.125ug/well. 1mg/ ml Pg-IgY could inhibit growth of P. gingivals. Conclusion The production and purity of IgY were high,and has strong specificity.The IgY had a combination capacity of specificity for P. gingivals andagglutination of P. gingivals on erythrocyte could inhibit by Pg-IgY. |
