Objective:To investigate the effect of metforminon proliferation and cell cycle of HEC-1A endometrial cancer cells of LKB1 gene overexpression.Methods:HEC-1A cells of LKB1 gene overexpression and control HEC-1A cells were treated by different concentrations of metformin.The proliferation rate, cell migration ability and cell cycle ofHEC-1 A cells was detected by colony formation assay, Trans well experiment and flow cytometer.The expression levels of LKB1 and mTOR mRNA and proteinin HEC-1A cells were detected by RT-PCR and western-blot.Results:compared with control, incubation with metformin inhibited a dose-dependent colonies, cell motility, percentage at S phase and expression of mTOR mRNA andprotein in HEC-1 A cells of LKB1 overexpression, respectively (P<0.05). But show promotion a dose-dependent percentage at G0/G1 phase and expression of LKB1 mRNA and protein, respectively (P<0.05)Conclusion:The proliferation rate and cell migrationability of HEC-1A endometrial cancercells of LKB 1 gene over expression inhibited by metformin. And the antiproliferative ability of metformin to endometrial cancer cells may bethrough LKB1/mTOR signaling pathway.
