Objective:To explore the general security of betamethasone microspheres-chitosan thermosensitive hydrogel complex delivery system as the SD rat knee injection,to explore the slow-release function of betamethasone microspheres-chitosan thermosensitive hydrogel complex delivery system in the joint local tissue of SD rat knee and to explore the therapeutic effects of betamethasone microspheres-chitosan thermosensitive hydrogel complex delivery system in the OA-type SD rat.Methods:(1)Taking healthy SD female rats,and left knee joints were injected by betamethasone injection, betamethasone microspher einjection of treatment dose and three different concentrations (7.37,12.82,17.58mg/mL, the concentration of microspheres)complex delivery system injection. Observating and scoring the left knee cartilage and synovium for gross and histological of each group mainly, to explore the general security of the complex delivery system(2)Taking healthy SD female rats,and left knee joints were injected by betamethasone injection, betamethasone microspher einjection and complex delivery system injection.Measuring the content of betamethasone in plasma, synovial fluid and synovial tissue fluid at different time points,and to explore the slow-release function of complex delivery system.(3)Taking healthy SD female rats,and their left knees were injected by mixture of papain,which became OA-type SD rats at week 4 post-injection.The OA-type SD rats’ left knee joints were injected by betamethasone injection, betamethasone microspher einjection and complex delivery system injection,which were killed within four weeks,and observating and scoring the left knee cartilage and synovium for gross and histological of each group mainly,to explore the efficacy of complex delivery system for knee OA.Results:Healthy SD rats left knee joints were injected by three different concentrations (7.37,12.82,17.58mg/mL, the concentration of microspheres)complex delivery system injection,and the functions of the rats’ major organs and tissues were normal within four weeks,in which the significant damage was not found.The complex delivery system injection has a good safety;The complex delivery system has a good slow-release function in SD rat left knee;The complex delivery system injection has a good efficacy for knee OA of SD female rats.Conclusion:The betamethasone microspheres-chitosan thermosensitive hydrogel complex delivery system has a good safety, a good slow-release function and a significant efficacy for knee OA of SD female rats through SD female rats in vivo.To study new forms of drug for OA treatment,the experiment provides a detailed theoretical basis.
