Objective:To investigate the relationship between HBV Pre-S region mutations and HBV-related HCC and to establish pre-s gene consensus sequence of HBV in Fusui Guangxi.Methods:According to case-control study,53 HBV-related HCC patients and 95 asymptomatic HBV carriers were selected from HCC high-risk cohort of Fusui. After extracting HBV DNA from sera, Pre-S/S region was amplified with PCR and directly secquenced. Nucleotide sequences in Pre-S region were import to MEGA6 for multiple sequence alignment by Clusta W program, After the alignment, a nucleotide with the highest frequency at each site in the Pre-S region of HBV genotype B or genotype C was termed as a nucleotide of consensus sequence.Results:Among the HBV-related HCC group and control group,41.5%(23 /53) and 10.5%(10/95) respectively had Pre-S deletions, the difference was statistical significant (χ2=19.271,P=0.000,OR=6.032).Among the HBV-relate d HCC group and control group,52.88%(28/53) and 37.9%(36/95) respecti vely had C3026T mutaion, the difference was not statistical significant(χ2 =3.092, P=0.079,OR=1.836). However, Among the HBV-related HCC gr oup with genotype C and control group with genotype C,71.8%(28/39) a nd 51.4%(36/70) respectively had C3026T mutaion, the difference was stat istical significant was significant(χ2=4.186, P=0.041,OR=2.404). Among the HBV-related HCC group and control group,32.1%(17/53) and 8.4%(8/ 95) respectively had Pre-S2 start codon mutation, the difference was statis tical significant was significant(χ2=13.559, P=0.000,OR=5.135). Among the HBV-related HCC group and control group,24.5%(13/53) and 5.3%(5/ 95) respectively had T53C mutaion, the difference was significant was st atistical significant(x2=11.819, P=0.001,OR=5.850). The Logistic regressi on analysis found that Sex(OR=2.907,95%CI 1.126-7.506), Pre-S deletion s(OR=7.021,95%CI 2.692-18.314),Pre-S2 start codon mutation(OR=5.633,9 5%CI 1.869-16.983) and T53C mutation(OR=5.313,95%CI 1.518-18.600) w ere associated with HCC. Stratification analysis showed that the prevalence of T53C mutaion was regard as interlayer heterogeneity after stratifying by sex. Pre-s gene consensus sequence of HBV genotype B and C were established in this study,the heterogeneity between the two genotypes wer e12.1%.Conclusions:Pre-S deletions,re-S2 start codon mutationsand and T53C mutaion were independently associated with HCC.Gender was a modified factor to T53C mutaion. The elect consensus sequence would impact the flnal results owing to the heterogeneityof epidemic strain in different territory,Therefore establishing the consensus sequence of HBV Pre-S region in Fusui is a fundamental work for HBV variation studies.
