| Objective:To investigate the role of ghrelin/ peptide YY / Agouti-related protein(AGRP) after the operation of gastrointestinal bypass in type 2 diabetes.Methods:Thirty adult SD male rats were selected, and 20 Of them successfuly were made as type 2 diabetic model. Ten of which were choosed as the sham-operation group(SO group, n=10), the other 10 rats as the Roux-en-Ygastric bypass group(RYGB group, n=10). The rest 10 of 30 rats are defined as the blank control group(BC group, n=10). The rats of the RYGB group were operated by RYGB. The rats of the SO group were operated by Laparotomy surgery.The diversification of ghrelin was tested by ELISA. The Blood Sugar(BS), triglycerides(TG), cholesterol(CHOL) of rats also were detected. After 4 weeks all rats were killed to test the level of PYY( ileal mucosa) and AGRP mRNA(hypothalamus).Results:1. Twenty SD rats were successfully made to be the diabetes models, and which Weight, diet, water intake were significantly more than that of the SD group. In plasma BS, TG and CHOL of the SO group and the RYGB group were significantly higher than that of the BC group (P<0.05). Expression of ghrelin of the SO group and the RYGB group were significantly higher than that of the BC group (P<0.05);2. There was no changes in weight, food and water intake between of the SO group and in the RYGB group. Thelevel of BS, TG and CHOL is significantly higher than that of the BC group and the RYGB group (P<0.05). These data in the RYGB group were lower than preoperation as the BC group (P<0.05).3. After 4 weeks postoperation, the level of PYY in Ileal mucosa of the RYGB group showed higher than that of the SO group (P<0.05). The expression of the ghrelin in the RYGB group is lower than preopration and approached to the BC group (P< 0.05). The AGRP mRNA of the SO group is Obviously higher thanthat of the RYGB and BC group (P<0.05)Conclusion:Gastrointestinal bypass operation can treat type 2 diabetes disease effectively. Weight and energy intake of type 2 diabetic rats changed to be nomal after operation and the level of BS, triglycerides and total cholesterol were significantly lower than preopration. The expression of the ghrelin, PYY and AGRP were different in the different groups. Ghrelin may be regulate by PYY and AGRP in type 2 diabetes. |
