Objectives Detect the expressions of CD55 and CD59 in the peripheral blood of anemia diseases and do relevant experimental examinations to investigate the clinical significance. Methods The expressions of CD55 and CD59 of 129 anemia patients selected in our hospital from June 2012 to February 2014 were detected with flow cytometry. 25 healthy persons were selected as observation group and FLAER test, Ham test, Rous test, and Reticulocyte counts were simultaneously done in some patients. Then the inspection results of the two groups were analyzed and compared,comprehensively. Results The CD55 and CD59 expressions in the peripheral erythrocytes and granulocytes of AA-PNH and PNH patients were markedly lower than those in the healthy group and the other anemia groups(P<0.05),whose expressions were less than 95.0%. The CD55 and CD59 expressions in some aplastic anemia and some myelodysplastic syndrome patients were also low, but higher than 48.0%. There was no statistically significant difference compared with the healthy group and some anemia groups(except AA-PNH and PNH groups,P>0.05). The CD55 and CD59 expressions in Pure red aplastic anemia、megaloblastic anemia、autoimmune hemolytic anemia and iron deficiency anemia patients.were in normal range.FLAER tests were done in some AA-PNH and PNH patients. The results showed that the expressions of PNH clones in the peripheral granulocytes and monocytes were markedly higher than observation group(P<0.05). For AA-PNH, PNH patients, the results between neutrophil PNH clones from FLAER tests and their CD55、CD59 negative expressions in the peripheral erythrocytes and granulocytes were significantly different(P<0.05),the results between monocyte PNH clones from FLAER testsand their CD55 、 CD59 negative expressions in the peripheral erythrocytes and granulocytes were also significantly different(P<0.05).Conclusions Detection of CD55、CD59 and FLAER test in the diagnosis of AA – PNH and PNH are very important. Up till now, they are the most sensitive and reliable methods in the diagnosis of PNH, and they also can be used as means to evaluate the curative effects and to observe the disease situation changes. There are important significances in the differential diagnosis of PNH and other anemia diseases.
