Objective:Peripheral blood stem cell were a hematopoietic cell population with a high degree of self-renewal and differentiation potential, which present in the peripheral blood,Under physiological conditions,hematopoietic stem cells in peripheral blood content was low, but donor was administered to he mobilization agent, the content of hematopoietic stem cell in peripheral blood was increased,CD34+cell in PBMNC was 0.5%-1.5%.With the collection of peripheral blood stem cell was simple, needs less time,and Recipient’s hematopoietic recovery and immune recovery was rapidly. Peripheral blood stem cell replace the bone marrow hematopoietic stem cells for clinical use gradually. Peripheral blood stem cell can promote hematopoietic, immune reconstitution and anti-tumor function etc. It has Confirmed that embryonic stem cells, cord blood stem cells, mesenchymal stem cells can release microvesicles,and carry the parental cells’Composition and properties, and without some drawbacks of the parental cells. There are no reports confirmed that if peripheral blood stem cell can release microcesicles in domestic and foreign.The purpose of this study is to explore that If peripheral blood hematopoietic stem cells release microvesicles,and if so, preliminary study the biological function of microvesicles.Content:1. If peripheral blood hematopoietic stem cells release microvesicles; 2.1f so, begin to characterize their biologic activity.Methods:1.The isolation of "Substance" in peripheral blood hematopoietic stem cells’culture supernatant after incubation,and if the "Substance" was microvesicles by using Electron microscopy;2.If peripheral blood hematopoietic stem cells release microvesicles,MVs were labeled with a PKH26 red florescent labeling kit and PBMNC was labeled with Hoechst 33258,and then viewed that if micro vesicles could enter PBMNC by confocal microscopy3.If so,begin to characterize that if microvesicles can change PBMNC’s Killing effect.Results:1. peripheral blood hematopoietic stem cell can release microvesicles;2.Peripheral blood hematopoietic stem cell’s microvesicles can enter PBMNC;3.Validated the microvesicles alter the receptor cells(PBMNC) Killing effect.Conclusions:1.we first confirmed that peripheral blood hematopoietic stem cell can release microvesicles; 2.Preliminary study the microvesicles can change the receptor cells’biological function.
