Object: This study is to investigate the association between gene polymorphisms-1112 C/T in the promoter region of the IL13 gene, CYP3A4*1G and susceptibility to penicillin allergy.Method:A case-control method was used in the study. The samples were divided into two groups, the case group (n=211) and control group (n=220), and all matched for gender and age. Selecting clinical 211 patients with allergic reaction to penicillin and 220 cases of healthy control in Han population of Gansu area to do the research, using the method of restriction fragment length polymorphism polymerase chain reaction PCR-RFLP to test the genetic distribution of allele and genotype frequencies of CYP3A4*1G and IL-13 -1112C/T, Hardy-Weinberg test was used later to confirm the group representative, software of SPSS 19.0 was used to perform a difference detection between the case and control group, thus to analyse the association.Result:The distributions of CC, CT and TT genotypes and C, T alleles in IL-13-1112C/T between the case and control group was statistically significantly different (P=0.006). And according to gender stratified statistics, the role of T alleles in male especially evident (P=0.002, OR=2.066,95%CI=1.297-3.289). No statistically significantly difference was found between the case and control group on the CC, CT and TT genotypes and C, T alleles in CYP3A4*1G site (P=0.333). There was still no significant difference even after a gender stratification.Conclusion: IL-13 -1112C/T gene may be a susceptible gene of penicillin allergic reactions, especially in male patients.
