Objective:The research is aim to investigating the effects of Bu-shen-huo-xie prescription, in the way of observing the clinical outcomes of the distal tibia fracture patience by taking herbal medicine after the procedure. Meanwhile, Animal experiments are used in exploring the effects of the migration of MSCs and the expression of Chemokine receptors in the fracture site. All these ways are to provide the accurate and reliable laboratory data of Chinese theory in treating factures.Methods:Clinical observation40 patience were randomly selected and divided into 2 groups, treatment and control ones. The method for treatment group is to taking the Bu-shen-huo-xie prescription after the procedures, as for the control ones, Jie-gu-qi-li tablet is given to. During the treatments, VAS score, Bone fracture healing time, Johner-Wurh’s tibiofibula score were used as the parameters in evaluation.Laboratory studiesThe rat femur fracture model was conducted and treated with Bu-shen-huo-xie prescription. Then the MSCs in the fracture site was isolated and cultured at 7th day,14th day and 21st day, respectively. Transwell assay was applied to examine the migration of 3rd passage MSCs, which is influenced by Bu-shen-huo-xie prescription. To clarify its mechanisms of promoting migration of MSCs by Bu-shen-huo-xie prescription, MSCs was tested by Western Blot and QPCR in protein and mRNA of its receptors CXCR4.Results:Bu-shen-huo-xie prescription plays a positive role in relieving pain、 promoting bony callus grow and shorting the fracture healing time. The results of Transwell assay revealed that Bu-shen-huo-xie prescription could promote migration of MSCs significantly at 7th.,14th day, compared with the model group and control group were significantly different, with statistical significance. After the 21st day, the migration of MSCs in the fracture site become weak gradually, compared with the model group were insignificantly different, with no statistical significance. The results of Western Blot、QPCR revealed that the expression of protein and mRNA of MSCs’receptors in fracture site are significantly upregulated at 7th、14th day, compared with the model group and control group were significantly different, with statistical significance.Conclusion:Bu-shen-huo-xie prescription could promote the fracture healing and improve the clinical outcomes of distal tibial fracture patience. And its mechanism may be associated with the migration of MSCs and the expression of its receptor CXCR4 in the fracture site.
