Objective:In this study,98 batches of samples from different sources of tangerine peel as the research object. By means of near infrared spectroscopy, UPLC fingerprint technology, UPLC/Q-TOF-MS technology, UPLC determination of the content, and a variety of statistical methods of cluster analysis, principal component analysis, consistency test, to carry out the evaluation on the quality of dried tangerine peel pure digital control index of Pericarpium Citri Reticulatae, respectively from the two aspects of character and chemical composition, in order to reveal the connotation of the genuineness of Pericarpium Citri Reticulatae. It is providing a new idea and play an exemplary role in the original study and quality evaluation of Pericarpium Citri Reticulatae, which have important implications to guarantee the quality and realize modernization of Traditional Chinese Medicine.Methods:In this study,98 batches of samples from different sources of tangerine peel were collected. (1) The near infrared spectra of tangerine peel from different sources. Using 20 batches of Pericarpium Citri Reticulatae as calibration set which to establish cluster analysis model and consistency test model. Besides, we use 61 batches of other tangerine peel and 17 batches of Pericarpium Citri Reticulatae as validation, in order to verify the accuracy of two models. (2) To establish UPLC fingerprints in water-soluble constituents of tangerine peel, it is a reference to establish the fingerprint of 37 batches of Pericarpium Citri Reticulatae. According to the comparison result, it were compared using the established UPLC fingerprint similarity (S) by98 batches of samples from different sources of tangerine peel. (3) The use of UPLC/Q-TOF-MS for 12 UPLC of common peaks in fingerprint were identified. (4) With hesperidin, nobiletin and tangeretin as indexs to establish a method for determination of the content of UPLC, the comparison with different varieties of tangerine peel, and the difference between different storage time and different harvest time of the flavonoid content of Pericarpium Citri Reticulatae. With hesperidin as the index, calculate the relative correction factor of nobiletin and tangeretin, and using the relative correction factor calculation of content of nobiletin and orange peel, and compared with external standard method and "quantitative analysis of multi-components by single-marker (QAMs)" results.Results:(1) Through the analysis of near infrared spectral cluster analysis and consistency test of the "tangerine peel" and "Pericarpium Citri Reticulatae", established cluster analysis model of "Pericarpium Citri Reticulatae" and consistency test model, this method validation showed that its accuracy is well. Genuine differences can overall reflect the "Pericarpium Citri Reticulatae" and "tangerine peel", can for the rapid identification of "Pericarpium Citri Reticulatae" by this method. (2) It was investigated to establish the fingerprint of 98 batches of dried samples, according to the similarity(S) results, and combined with principal component analysis method, obtained Citrus reticulate "Chachi" and other varieties of Citrus tangerine peel water-soluble components have certain differences, and the difference can be used to identify the genuineness of "Pericarpium Citri Reticulatae" index. (3) The use of UPLC/Q-TOF-MS for the 12 common peaks in UPLC fingerprint spectra confirm of tangerine peel, and preliminary explanation of the fragmentation pattern by three main kinds flavonoid constituents of Pericarpium Citri Reticulatae in the extract. (4) Using the UPLC method for the determination in content of the determination for 98 batches tangerine peel with cluster analysis and principal component analysis. Analysis methods can reflect the genuine "Pericarpium Citri Reticulatae", besides compared the determination in content of flavonoids with different varieties, different storage time and different harvest time change. Meanwhile 98 batches of samples determined by QAMs method and external standard method, nobiletin and tangeretin were no significant difference.Conclusion:Based on near infrared spectroscopy technology, UPLC fingerprint, UPLC/Q-TOF-MS and determination of the external standard method and QAMs method, also can identify specific to wide tangerine peel, a new method is provided to reveal the genuineness connotation.
