Objective The paper aims to research how Didang Tang and its partial prescriptions influence the myocardial cell hypertrophy in lab rats with diabetes on the basis of JAK-STAT pathway.Methods The research needs 130 healthy male Sprague Dawley rats and each rat weighs 200~240g. Among them, 110 Sprague Dawley rats were intraperitoneally injected STZ in a dose of 55mg/kg to copy DM rats modules. After 72 hours STZ injection, the rats were measured random blood glucose by caudal vein blood drawing.The rats whose random blood glucose level >16.7mmol/L were regarded as successful DM rats module copies. Then, 100 successful DM module copies were divided into 5groups with 20 rats per group in random : the model group, the Chinese medicine Didang Tang group, the Peach seeds and rheum officinale group, the Leech and gadfly group, the Valsartan capsule group. Each Chinese medicine group were separately given Didang Tang, Peach seeds and rheum officinale, Leech and gadfly herbal decoction.The concentration of these herbal Chinese medicine in water is1g/ml. And rats in each Chinese medicine group were given intragastric administration in 10ml·Kg-1of the mixed liquid. The Western medicine Valsartan capsule group were given intragastric administration in 80mg/kg. The normal group and the module group were given distilled water with the same quantity. All the rats in above six groups were continually given intragastric administration once a day for8 weeks. We adopted western blot to detect the expression of AK2, ANP, STAT3 protein in rats’ myocardium tissue and adopted RT-PCR to detect the expression of JAK2、STAT3 m RNA in order to statistical analysis.Results(1)According to Western blot indication, compared with the normal group,ANP of rats in the model group was significantly up(P<0.01); compared with the model group, ANP of rats in both of the treating groups were low(P<0.01);compared with the two partial prescriptions group, ANP of rats in the Didang Tang group was significantly low(P<0.01); compared with the Valsartan capsule group,ANP of rats in the Didang Tang group was low, but P>0.05, the differences did not have statistical meaning.(2)According to Western blot indication,(JAK2:compared with the normal group, JAK2 of rats in the model group was significantly up(P<0.01); compared with the model group, JAK2 of rats in both of the treating groups were low(P<0.01);compared with the Valsartan capsule group, the Leech and gadfly group, the Peach seeds and rheum officinale group, JAK2 of rats in the Didang Tang group was significantly low(P<0.01).(STAT3: compared with the normal group, STAT3 of rats in the model group was significantly up(P<0.01); compared with the model group,STAT3 of rats in both of the Didang Tang groups, the Valsartan capsule group and the Peach seeds and rheum officinale group were significantly low(P<0.01), STAT3 of rats in the Leech and gadfly group was low two(P <0.05); compared with the Leech and gadfly group, STAT3 of rats in the the Didang Tang group was low(P<0.05), but compared with the Peach seeds and rheum officinale group, the Valsartan capsule group, STAT3 of rats in the the Didang Tang group was not significantly low(P >0.05).(3)According to Western blot indication,(JAK2:compared with the normal group, JAK2 of rats in the model group was significantly up(P<0.01); compared with the model group, JAK2 of rats in both of the treating groups were low(P<0.01);compared with the Didang Tang group, JAK2 of rats in the Leech and gadfly group was not significantly up(P >0.05), JAK2 of rats in the Peach seeds and rheum officinale group was low(P <0.05), JAK2 of rats in the Valsartan capsule group was significantly low(P<0.01).(STAT3: compared with the normal group, STAT3 of rats in the model group was significantly up(P<0.01); compared with the model group,STAT3 of rats in both of the treating groups were low(P<0.01); compared with the Didang Tang group, STAT3 of rats in both of the Valsartan capsule group and the Leech and gadfly group was not significantly low(P >0.05), STAT3 of rats in the Peach seeds and rheum officinale group was significantly low(P<0.01).Conclusion The abnormal key molecule of JAK-STAT signal transduction pathway is one of the important pathogenesis of diabetic myocardial cell hypertrophy. Didang Tang can regulate and control JAK-STAT cell signal transduction pathway, reduce the expression level of JAK2 and STAT3, prevent and cure diabetic myocardial cell hypertrophy.
