Objectives1. To investigate the correlation between aldehyde dehydrogenase2 (ALDH2) polymorphisms and aortic dissection (AD) in Chinese Han People.2. To determine the risk factors for AD and risk factors leading to hospital death.MethodsA case control study method was adopted.51 aortic dissection patients who came to Qilu Hospital of Shandong University from May 2014 to May 2015 and 51 cases with no aortic dissection, who were chosen according to the constitute ratio by age and gender of the patients group were involved in this study to analysis the influence of the ALDH2 polymorphisms to the incidence and risk factors for aortic dissection and the hospital death. The clinical base-line data were collected using medical records containing the subjects’clinical characteristies, including age, gender, smoking, drinking, hypertension, diabetes, D-Di, lipid items, clinical outcomes, et al. The lipid items included total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C). Blood samples were collected at the ulnar vein of 2 ml in tubes within 12 hrs after the patient accepted in. We extracted the DNA with a DNA extraction kit, then took the polymerase chain reaction (PCR) to extend DNA production. Then we purified the DNA production and definited the ALDH2 genotypes by the method of gene sequencing of the Boshang Shanghai Corporation. AD patients were divided into two subgroups according to their clinical outcome:dead group (patients died in hospital) and alive group (patients improved or transfered). Statistical analysis were performed using SPSS 20. After adjustment for confounding factors, a multivariate logistical regression analysis was performed to identify the odds radio and 95%CI. A p-value of less than 0.05 was considered statistically significant.Result1. The difference of the constituent ratio of age and gender, smoking of the two groups was not significant. The wild ALDH2 genotype, hypertension of the AD group were more or higher than that in the control group (P<0.05), while the lipid items, DM of the AD group were less or lower than that in the control group (P<0.05)2. After adjustment for smoking, drinking, hypertension, lipid items, a multi-variable logistic regression showed the wild genotype (ALDH2* 1/* 1) was an independent risk factor of the incidence of AD (OR=2.72,95%C1=1.16~6.41)3. D-Di and Type A aortic dissection of the death group were higher or more than that in improved group.Conclusions1. ALDH2 polymorphisms was an independent risk factor of aortic dissection in Chinese Han people.2. D-Di and Type A AD were more associated with hospital death of AD patients.
