Pirh2 is the new member of E3 ubiquitin ligase family,has intrinsic“Ubiquitylation”active RING-H2 structure domain;p53 is found that the highest correlation with human malignant tumor suppressor genes.Existing research shows that Pirh2 can ubiquitylate p53,lead to the degradation of p53,which involved in cancer development.However,The development and metastasis of cancer is closely related to the cancer cells invasion and metastasis ability.【Objective】This study explored Pirh2/p53 expression and significance of the breast cancer cell lines in different metastatic potentiality, and using the normal breast epithelial cell lines as a control,in order to prove that pirh2 and p53 was associated with the invasion and metastasis ability of breast cancer cells.Its could provide preliminary theoretical basis to find new molecular mechanisms and therapeutic targets to the diagnosis and treatment of breast cancer.【Methods】To cultive normal breast epithelial cell lines MCF-10 A,MCF-7 breast cancer cell lines with low metastatic potentiality,MDA-MB-231 breast cancer cell lines with moderate metastaticpotentiality,MDA-MB-435 s breast cancer cell lines with high metastatic potentiality;q PCR and Western blot methods were used to detect the expressions of Pirh2 and p53 in normal breast epithelial cell lines and breast cancer cell lines with different metastatic potentiality.【Results】1.Pirh2 and p53 had expressioned in normal breast epithelial cell lines MCF-10 A and MCF-7 breast cancer cell lines with low metastatic potentiality,MDA-MB-231 breast cancer cell lines with moderate metastatic potentiality,MDA-MB-435 s breast cancer cell lines with high metastatic potentiality.2.The expression of Pirh2 in breast cancer cell lines was higher than normal breast epithelial cell lines,and the difference was statistically significant(P<0.05).The expression of p53 in breast cancer cell lines was lower than normal breast epithelial cell lines,and the difference was statistically significant(P<0.05).3.With the increase of metastatic potentiality of breast cancer cell lines, the expression level of Pirh2 was on the rise, the difference was statistically significant in comparison between groups(P<0.05);p53 was on the fall, the difference was statistically significant in comparison between groups(P<0.05).4.The expression level of Pirh2 and p53 had significant negative correlation(rm RNA=-0.904,Pm RNA<0.01;rprotein=-0.953,Pprotein<0.01).【Conclusion】1.The expression of Pirh2 and p53 was associated with the invasion and metastasis ability of breast cancer cells.Pirh2 had a high expression in breast cancer cell lines,and p53 had a low expression in breast cancer cell lines.With the increase of metastatic potentiality of breast cancer cell lines,the expression level of Pirh2 was on the rise,and p53 was on the fall.2.The negative correlation of Pirh2 and p53 suggest that the exist of Pirh2/p53 cell signaling pathways in breast cancer and Pirh2 negative regulation of p53.
